Influence Of Astragalus And Notoginseng On Expression Of MMP₂ In AS Model Aorta

Objective: Through the observation on the effect of matrix metalloproteinase Ⅱ's (MMP2) expressing of atherosclerosis (AS) mouse model from Chinese herbs for replenishing the vital qi and and removing blood stasis Astragalus and Notoginseng, study the function of replenishing qi and removing blood stasis to AS and its mechanism. Method: Divide Wistar mouse randomly into normal group and model-making group. After feeding model group with high fat feed for three months, the blood fat of both group are tested .Take main artery to have pathological examination.Then, model group are divided randomly into six groups, normal group (A), model group (B), quercetin group (C), total Panax Notoginseng aponin group (D), quercetin and total Panax Notoginseng aponin low dosage group (E), quercetin and total Panax Notoginseng aponin high dosage group (F), chuanxiong Rhizome group (G).Take intervention for 4 weeks, take quantitative test of MMP2 with ELISA method and test CRP and blood fat. Result:(1)histological study shows the intema of B group is thicker, the inner elastic plate is broken frothy cells infiltrated, fat spot protrude inward, while as for the rest group, the intema is normal.(2)Compare the fat in blood of A group and B group before treatment: there's no obvious differences on HDL(P＞0.05);TC,TG,LDL are all higher than A group (P﹤0.001).(3)After treatment , compare C,D,E,F group respectively with B group,TG,LDL,CRP and MMP2of C,D,F,F group are all lower than that of B group (P﹤0.001), and F group is the most obvious;HDL of C,D,F,F group are higher than B group, especially the C group(P﹤0.001),and the rest(P﹤0.05);TC of D,E,F group are all lower than B group especially F group(P﹤0.001).(4)After treatment, to compare C﹑D﹑E﹑F,each two as a pair,TG,TC,LDL,CRP and MMP2 of F group are all the lowest. there's no obvious differences on HDL.(5)After treatment, to compare A,B,F,G,each two as a pair,TG,TC,LDL,CRP and MMP2 of F group are all the lowest and HDL of G group are highest. There's no difference between F,G group and A,G group; on TC,HDL,CRP between A,F,G group, on LDL and MMP2 between A and G group ,but there're differences between the rest groups. Conclusion:Both quercetin and total Panax Notoginseng aponin can resist AS. It's better to use them together than singly used. It is likely that there's some relationship with the expression of the inhibition of MMP2 and with the process of inhibition of the inflammation, the TG,TC,LDL are lowered, and HDL is raised.
