| ObjectiveTo investigate the therapeutic effect of PNS on apolipoprotein E gene knockout(Apo E-/-)inflammatory response induced by high-fat diet(basal feed,fat,cholesterol ratio 84.75:15:0.25)in mice with atherosclerosis(AS)and discuss its mechanism.Methods24 Apo E-/-mice were randomly divided into model group,according to the weight PNS low dose group(60 mg/kg)and PNS high dose group(180 mg/kg),each group of eight,high-fat feed(basic feed,fat,cholesterol ratio is84.75:15:0.25)feeding 8 weeks make AS model,8 wild type C57BL/6 j mice AS normal control group,the average feed,PNS low and high dose group according to the dose to give 0.2 ml,normal control group and model group to fill the stomach every day the same amount of 0.5%CMC Na,continuous feeding for 8 weeks.The serum levels of TG,TC,LDL-C and HDL-C were measured by biochemical analyzer.HE staining was used to observe the pathomorphological changes of the aortic root of mice in each group.Lipid deposition in aortic roots of mice in each group was observed by oil red O staining.Expression of IL-1βand TNF-αin serum was detected by ELISA.The levels of NF-κB,Il-1β,Il-6,TNF-αand Calpain1 in the aortic root tissues were measured by Western Blot.ResultsCompared with normal control group,model group mice serum TG levels rise,TC,LDL-C levels were significantly increased(P<0.01),HDL-C levels drop,aortic root tissue in mice irregular arrangement,the nucleus gathered,aortic root organization positive staining cells was significantly increased,has obvious plaque lipid deposition,can be clearly observed apparent dot patches to the lumen,lumen narrowing obviously,aortic root tissues in mice NF-κB,IL-6,IL-1β,TNF-αand Calpain1 protein expression were significantly higher(P<0.01);Compared with model group,PNS can decrease the mice serum TG,TC,LDL-C levels,increase HDL-C levels,PNS to arrange normal aortic root tissues in the medicine group,cell morphological recovered,reduce lipid deposition in the aortic root organization phenomenon,aortic root tissues NF-κB,IL-6,IL-1β,TNF-αand Calpain1 protein expression were significantly reduced.ConclusionPNS can regulate blood lipid levels,reduce inflammation in aortic root tissues,and reduce lipid plaque deposition in atherosclerotic mice induced by high-fat diet,which may be related to down-regulation of the NF-κB pathway. |
