Sixteen rats were randomly chosed from seventy-two femalesprague-daweley (SD) rats aged 10 month which were equally divided into group of sham operation with normal physiological saline A and B. The left fifty-six rats were randomly divided into 7 groups 3 days after ovariectomy: groups of osteoporotic animal model with normal physiological saline A and B, preventing groups of osteoporotic animal model with low dosage and high dosage of BBC, treating groups of osteoporotic animal model with low dosage and high dosage of BBC, treating group of osteoporotic animal model with QIANGGU capsule. Each group included 8 rats. Ovariectomy was used to establish the osteoporotic animal model and the total body bone mineral density (BMD) of rats were detected in 12 weeks and 24 weeks after operation. The blood from the rat's left ventricle was centrifugated to get the blood serum. The concentrations of serum parathyroid hormone (PTH)and calcitonin (CT) were detected by radioimimunoassay(RIA). The expression of Type I collagen on protein level in bone tissue was detected by immunohistochemistry method.2. Twelve rats were randomly chosed from forty-two female SD rats aged 10 month which were equally divided into group of sham operation with normal physiological saline A and B. The left thirty rats were randomly divided into 5 groups 3 days after ovariectomy: groups of osteoporotic animal model with normal physiological saline A and B, preventing groups of osteoporotic animal model with BBC, treating groups of osteoporotic animal model with BBC, treating group of osteoporotic animal model with QIANGGU capsule. Each group included 6 rats. Ovariectomy was used to establish the osteoporotic animal model and the expressionof Type I collagen mRNA on gene level in bone tissue was detected by real time fluorescence quantitative PCR(FQ-PCR) method.3. The differences between groups were compared by ANOVA in SPSS11.0.Results: 1. The status of osteoporosis in ovariectomy groups was confirmed by total body BMD measurement 12 weeks after ovariectomy. The results of the total body BMD in 12 and 24 weeks after ovariectomy of preventing groups with high and low dosage of BBC were all higher than groups A of osteoporotic animal model (P<0.05), which were all close to groups A of sham operation(P>0.05). The results of the total body BMD in 24 weeks after ovariectomy of treating groups of osteoporotic animal model with high and low dosage of BBC were all higher than groups B of osteoporotic animal model (P<0.05), in which the effects of high dosage for treating groups with BBC were close to groups B of sham operation(P>0.05), but the effects of treating group with low dosage of BBC and treating group with QIANGGU capsule were both lower than groups B of sham operation (P<0.05).2. The level of CT in serum in 24 weeks after ovariectomy of preventing and treatin groups with high and low dosage of BBC were all higher than groups of osteoporotic animal model (P<0.05), in which the effects of preventing and treating groups with high dosage of BBC were close to groups of sham operation(P>0.05), but the effects of preventing and treating group with low dosage of BBC and treating group with QIANGGU capsule were lower than groups of sham operation (P<0.05).3. The level of PTH in serum in 24 weeks after ovariectomy of preventing and treatin groups with high and low dosage of BBC were all lower than groups of osteoporotic animal model (P<0.05), in which the effects of preventing and treating groups with high dosage of BBC were close to groups of sham operation(P>0.05), but the effects of preventing and treating group with low dosage of BBC and treating group with QIANGGU capsule were higher than groups of sham operation (P<0.05).4.
