Objective: To study effects of Benefiting-bone Capsule (BBC) on cancellous structure of osteoporosis model ovariotomy rats by bone mineral density and quantitative analysis of bone histomorphometry parameter and star volume.Methods: The seventy-two 10-month female Sprague-daweley rats were randomly divided into treatment part (sham, model, BBC treatment high, BBC treatment low dose and QIANGGU CAPSULE control groups) and preventing part (sham, model, BBC preventing high and BBC preventing low dose grougs).All rats were taken ovariotomy except rats in sham group. The rats in treatment groups were given herbs after 12 weeks. The rats in preventing groups were given herbs after 3 days. All rats were checked bone mineral density by Duel Energy X Ray Asorptiometry at 12th week and 24th week. After 24 weeks, the bones of all rats were taken and cut into slices. Double fluorescence marks were taken before taking bones. All slices were study by quantitative analysis of bone histomorphometry parameter and star volume.Results: The general bone mineral density, trabecular area, trabecular plate thickness, trabecular number and trabecular nodes of rats in model groups were lower obviously than that in sham group, and free-ending trabecular, trabecular plate separation and star volume was higher obviously than that in sham group.In BBC treatment groups, QIANGGU CAPSULE control group and BBC preventing groups, the general bone mineral density, trabecular area, trabecular plate thickness, trabecular number and trabecular nodes of rats was higher obviously than that in model grop, and free-ending trabecular, trabecular plate separation and star volume was lower obviously than that in model group. Their difference had significance markedly(P<0.05 or P<0.01). The parameter in BBC high dose group were closed with that in preventing groups, and their difference hadn't statistics significance (P>0.05). Mineral aggradation rate in treatment and preventing groups were lower than that in model group, and higher obviously than that in sham group. Treatment low dose group and control group compared respectively with model group, and treatment low dose group and preventing groups compared respectively with sham group. Their difference of mineral aggradation rates was all had statistics significance ((P<0.05). Contrasted among groups given herbs at the effect of preventing and treatment, BBC treatment high dose group was superior to BBC treatment low dose group and control group, and BBC preventing high dose group was superior to BBC preventing low dose group.Conclusions:It was suggested that osteoporosis model rats could been copied succeedingly by ovariotomy. BBC could heighten general bone mineral density of model rats, and improve trabecular structure, and increase trabecular area, trabecular plate thickness, trabecular number and trabecular nodes, and drop trabecular plate separation, and decrease free-ending trabecular, and shorten mineral aggradation rat, and reduce star volune and space among trabeculars.
