Objective:This study investigated the expression of Vascular endothelial growth factor(VEGF) and P-glycoprotein (Pgp) in colorectal carcinoma and evaluated the roles of them in development,invasion and metastasis of colorectal carcinoma as well as their interaction in order to provide theoretical foundation for clinical therapy.Provide the basis for evaluation of prognosis,reversing multidrug resistence and targeting gene therapy.Materials and methods:56 cases of surgically resected colorectal carcinoma samples and 22 cases of well collected.All the tissues were fixed in 10% neutral formalin and embedded in paraffin.Streptavidin/Peroxidase(SP) technique was used to examine the expression of VEGF and Pgp in colorectal carcinoma and in non-tumor colorectal samples. x2 -test was used to analyze the date.There is a statistical significance when P<0.05.Results: The positive staining of VEGF is mainly located in cytoplasm. There was positive staining in some membrane and ECM.In colorectal carcinoma and in non-tumor colorectal samples,the positive rates of VEGF were 64.3% and 27.3% respectively.There were significant differences(P<0.05).In the well differentiated group,moderately differentiated group and poor differentiated group,the positive rates of VEGF were 43.8ï¼…(7/16),61.9ï¼…(13/21)and 84.2ï¼…(16/19)respectively.There were significant differences(P<0.05)when the expression of these three group were compared with each other. Of 56 cases colorectal carcinoma,in the group without serosa infiltration and the group with serosa infiltration,the positive rates of VEGF were 41.7%(10/24),81.3%(26/32).There were significant differences(P<0.05) between the two groups.In the group with lymph node metastasis and the group without lymph node metastasis,the positive rates of VEGF were 80.6%(25/31) and 44.0%(11/25).There were also significant differences(P<0.05) . The positive staining of Pgp is located in both cytoplasm and ECM. In colorectal carcinoma and in non-tumor colorectal samples, the positive rates of Pgp were 46.4% and 36.4% respectively. In the well differentiated group, moderately differentiated group and poor differentiated group, the positive rates of Pgp were 43.7%(7/16),38.1%(8/21),57.9%(11/19) respectively. There were no significant differences(P>0.05)when the expression of these three grouped were compared with each other. Of 56 cases colorectal carcinoma,in the group without serosa infiltration and the group with serosa infiltration,the positive rates of VEGF were 45.8%(11/24),46.9%(25/32).There were no significant differences(P>0.05) between the two groups.In the group with lymph node metastasis and the group without lymph node metastasis,the positive rates of VEGF were 61.3%(19/31) and 28.0%(7/25). There were significant differences(P<0.05) . Of 56 cases colorectalcancer,21 cases were positive expression of both VEGF and Pgp while 15 cases were negative expression of both VEGF and Pgp.There was close correlation between VEGF and Pgp expression.The expression of VEGF and Pgp has no correlation with the patients'age,sex and tumor size(P>0.05). Conclusion: The positive rates of VEGF and Pgp gradually increased with the progression of colorectal carcinoma. It is demonstrated that VEGF and Pgp have close relationship with the carcinogenesis and the development of colorectal. The expression of Pgp in carcinoma not benefit to carry through chemotherapy after resection but the expression of Pgp in non-tumor colorectal has some protection in the later therapy.The overexpression of VEGF was correlated with differentiation, infiltration and lymph node metastasis. The overexpression of Pgp was correlated with lymph node metastasis. It was indicated that the overexpression of them might occur in the late phase of colorectal carcinoma progression and they had closely correlation with the invasion and metastasis of colorectal carcinoma.It is suggested that the positive expression of VEGF and Pgp might be of value in guiding clinical treatment and in judging the prognosis of colorectal car...
