Objective : To evaluate the therapeutic effect of bFGFand GbE on acute cerebral infarction with medical imaging ; toinstruct the clinical application of neuroprotective agentcombined with herbal medicine . Methods : 40 healthy adult cats , each weighting3.5±0.2kg , were equally devided into 4 groups randomly . Allanimals were operated to establish the feline model of cerebralfocal ischemia-reperfusion by the approach of left transorbital .They were injected in vein with sodiumchloride for group A ,bFGF for group B , GbE for group C , bFGF+GbE for group Drespectively for 3 hours once a day . MR scans were performedjust after the operation , including T1WI , T2WI , FLAIR ,DWI , PWI . All scan sequences were repeated 2h , 6h , 12h ,48h ,72h , 5d ,7d after the operation . Philips' criteria score wasused to grade them 7 days after the operation . All animals weresacrificed by intracardiac injection of 2.5% formaldehydum plus1.5% glutaraldehyde , and the perfusion fixed brain wasremoved by craniectomy . Specimens were selected based onthe abnormal manifestation on MR images , stained with HE orimmunohistochemistry , and examined by light microscope and 4英 文 摘 要electron microscope . Result : The neurological defects score in Group A isremarkably higher than that in Group B , C and D . MR imaging showed the region of abnormal high intensityin area of MCA in DWI right after the reperfusion , associatedwith the decrease of the ADC value , and the other scansequences such as T1WI , T2WI , FLAIR can not detect theabnormal signal at the same time . 2 hours later , T1WI , T2WI ,FLAIR began to imagine the area and scale of the lesion clearly ,and showed the extensive abnormal signal . 12 hours later , thelesion developed more seriously , and the signal became muchhigher than before . 24 hours later , the edema around the lesionreduced significiantly and the border of the lesion appearedclearly . In each period , the control group (Group A) providedmore serious imagines than other groups .By learning MRimagines of all groups , a relatively fixed area was found inADC images right after the reperfusion , which provided aextremely low ADC value . Compared with the pathology , itcould be proved to be infarction area , and around it was thepenumbra . Through comparing the infarction volume rate withADC images and T2WI images 7 days later , the penumbra areacould be assured . The study shows this area of bFGF plus Gbegroup is much less than that of other groups . Morphological changes : Under LM , in bFGF plus GbEgroup , neurons appeared slightly degenerate , and the quantityof survival and normal nerve cells was relatively high . Under 5英 文 摘 要EM , the normal morpha of neurons and gliocytes remained ,and edema of intra- and extracell disappeared .Immunohischemisty also showed in this group the calculation ofpositive cells was high . Statistic results : On all variables including apoptotic cells ,MAP , the neurological defects score and infarction volume ,bFGF plus GbE group had significiant difference compared withthe control group and bFGF group ,the GbE group (P<0.05).The infarction volume measured on T2WI before sacrifice , theneurology defect score , the calculation of positive MAP cellsand the apoptosis cells within 10 regions of interest of differentgroups were all analysed by factorial design (SPSS 11.5). Conclusion : The feline model of cerebral focalischemia-reperfusion we established has excellent stability ,simple operation and reproducibility and is suitable for medicalimaging study . This study proves that both bFGF and GbE canenhance each other's therapeutic effects .
