AIM: The purpose is to research on the relationship of basic fibroblast growth factor (bFGF)and silicosis, clarify the effect of bFGF on the formation of fibrosis in the level of cells andmolecules, and detect the expression and location of bFGF in silicosis.METHODS: MTT was utilized to detect the role of the bronchoalveolar lavage fluid(BALF) of the mice exposed to silica in promoting proliferation of 3T3 cells. Conditionedmedium with bFGF-neutralizing antibody was used to treat cultured mouse lung fibroblastsgrowth under identical condition as described. The production of collagen was examined byMasson staining. The Expression of bFGF was detected by immuno- histochemicaltechnique.RESULTS: B ALF from model animals up-regulate the growth of lung fibroblast ( P<0.01vs control group ) and anti-bFGF antibody decrease its effect dramatically (P<0.01 vsantibody+21dBALF group; P>0.05 vs control group) . The production level of collagen inmodel animals of silicosis was remarkably higher than control group. The bFGF high-levelexpressed in lung macrophage.CONCLUSION: BALF of silicosis model of animals evidently promotes proliferation oflung fibroblast and the anti-bFGF antibody can restrain its effect. It indicates that the bFGFis high-level expressed in silicosis model of animals with the overproduction of collagenwhich participates the development of silicosis.
