Objective The purpose of this study was to investigate the change of NF-κB in the rat heart following acute myocardial ischemia evoked by coronary artery occlusion(CAO) and to investigate the intervention effects of morphine and tramadol on NF-κB. Methods Male Sprague-Dawley rats were randomly divided into group C,the control group,with non-CAO; group CAO30min,with CAO for 30 minutes; group CAO1h,with CAO for 1 hour; and group CAO3h, with CAO for 3 hours; group CAO6h,with CAO for 6hours; group M,with Morphine+CAO6h; and Group T, with Tramadol+CAO6h. Coronary artery was occluded except in group C. Rats were pre-administrated 1.25mg/kg morphine in group M and 12.5mg/kg tramadol in group T through caudal vein before CAO 15 minutes, respectively. Then rats were killed at different time points, and the heart was dissected in order to perform in immunohistochemistry (IH) and Flow cytometry(FCM) of heart tissue, the NF-κB of heart tissue were examined and analyzed semi-quantitively for its variation by using IH technique with IDA-2000 digital micro-image analysis systems and using FCM technique. Results The expression and activation of NF-κB increased significantly at 30min,1h,3h,6h after CAO as compared with control group(p<0.05), and there was an increasing trend with development of myocardial ischemia, But there was no significant difference in activation of NF-κB between CAO1h and CAO3h, then the level of NF-κB was much higher at CAO6h than that of other group. The expression and activation of NF-κB decreased significantly in group M and group T, But there was no significant difference between group M and group T. Conclusion Myocardial ischemia alone can activate the NF-κB in myocardial tissue, and NF-κB might play an important role in the process of myocardial ischemia. Morphine and Tramadol decreased the expression and activation of NF-κB in myocardial tissue and might play an important myocardial preservation role during acute myocardial ischemia.
