Objective: The purpose of this study was to explore the possible roles of TNF-αand SP in the neurogenic mechanisms of myocardial injury induced by coronary artey occlusion, the potential mechanisms of acute local myocardial ischemia provoking globle injuries of the heart. Accordingly the surveys were made about the temporal and spatial expressions of TNF-αand SP in myocardiums, dorsal root ganglion and spinal cord of rats after the coronary artery occlusion. Effects of pre-treatment with morphine, tramadol, rigitine, esmolol, and atropine on the expression of TNF-αand SP were investigated.Methods: The experiment was carried out in two steps. In step one, adult male SD rats were randomly devided into three groups: Control group (group C), the samples were harvested from normal healthy rats directly after anaesthesia; Sham group (group S), with the same scheme of surgery with non-CAO; Coronary artery occlusion group (group CAO). Then group S and group CAO were devided into eight subgroups: group CAO0.5h, with CAO for 30 minutes; group CAO1h, CAO3h, CAO6h, CAO1day, CAO2day, CAO3day and CAO7day, with CAO for 1 hour, 3 hours, 6 hours, 1day, 2day, 3day and 7day respectively. Then rats were killed at different time points, and the samples were collected for immunohistochemistry (IHC) and ELISA. The observations were made about the temporal and spatial expressions of TNF-αand SP in myocardiums, dorsal root ganglion (DRG) and spinal cord of the rats after the coronary artery occlusion.In step two of the experiment, the time point of highest expression of TNF-αand SP after CAO was chosen respectively for interventional study according to the results of step one experiment. Adult male SD rats were randomly devided into eight groups: Group C (Control group); Group S (Sham group); Group CAO (Coronary artery occlusion group); Group M (Morphine group); Group T (Tramadol group); Group R (Rigitine group); Group E (Esmolol group) and Group A (Atropine group). Rats were pre-treated with 1.25mg.kg-1 of morphine in group M; 12.5mg.kg-1 of tramadol in group T; 0.4mg.kg-1 of rigitine in group R; 3.0mg.kg-1 of esmolol in group E and 1.0mg.kg-1 of atropine in group A through caudal vein 15 minutes before CAO. Then rats were killed as scheduled, and the samples were collected in order to perform in immunohistochemistry (IHC), ELISA and Real Time RT-PCR.Results: (1) The expression of TNF-αin myocardium, DRG and spinal cord increased significantly after CAO as compared with the baseline in the control group(p<0.05).
