Experimental Study On The Effects Of Shouwudanshen Formula On Acute Myocardial Ischemia And Cardiac Hemodynamics In Narcose Dogs

Objective:The investigation aim directly at Coronary heart disease of Yinxuxueyu.The formula is made up of some traditional Chinese medicine,such as Shouwu and Danshen. To observe the effects of Shouwudanshen formula on Acute Myocardial Ischemia and Cardiac Hemodynamics in narcose dogs. Methods:The model of myocardiac ischemia in narcose dogs was established by ligating on left anterior descending branch of coronary artery. After injecting drugs by alimentary tract, the level ( E-ST) and range (N-ST) of myocardiac ischemia and the range of myocardial infarction was measured by epicardium electrocardiogram and histology(TTC staining). At that time, coronary artery blood flow, blood oxygen content of arteria and vein, Creatine kinase (CK), MB isoenzymes of creatine kinase (CK-MB), lactic dehydrogenase(LDH), endothelins(ET), thromboxane B2(TXB2) and 6-keton-prostaglandin FIO (6-KetoPGFia ) were measured; the effection of Hemodynamics of normal dogs with narcotized and opened thoracic cavity were observed such as left ventricle systolic pressure (LVSP) , left ventricle diastasis pressure (LVEDP), the max changing velocity of left ventricle pressure ( dp/dtmax ) , the odds of the max ascending and changing velocity of left ventricle pressure (+dp/dtmax/ LVP) ,heart rate (HR) , cardiac output (CO) and cardiac index (CI) etc. Result:Shouwudanshen formula can obviously decreased S-ST , the odds of range of myocardial infarction and heart or left ventricle and N-ST. It can increased coronary artery blood flow and decreased myocardium oxygen consumption.lt had the role of preventing LDH ,CK and CK-MB from step-up and preventing ET from releasing.lt also can accelerate 6-KetoPGFia to release and increase the odds of 6-KetoPGFio and TXBj. Shouwudanshen formula had the role of decreasing LVEDP and increasing CO,CI and the odds of +dp/dtmax and LVSP. Shouwudanshen formula is significantcompared with model group (P<0.05-0.001) . The action of Shouwudanshen formula connect with the dose. The high dose team have more important action, when narcose dogs haved used the drugs for 180 minutes, ?ST\ N-ST is decreased 51. 14 + 7. 61%, 20. 11+5. 58%; the odds of range of myocardial infarction and heart or left ventricle is droped 5. 5 + 22. 30%^ 8. 53+ 3. 19%; coronary artery blood flow increased 20.91 + 12. 33% ; myocardium oxygen consumption is decreased 6.49 + 8.99%; CO, CI and +dp/dtmM/ LVSP are increased 5. 74+16. 43%N 5. 74+ 16. 43%> 10. 86 + 20. 52%; Shouwudanshen formula is significant compared with model group (P<0.001) . Conclusion:Shouwudanshen formula had the role of increasing coronary artery blood flow, decreasing myocardiac damage and myocardium oxygen consumption, ameliorating myocardiac ischemia , cardiac function and endothelium function and became the therapy of Coronary heart disease. The high dose team of Shouwudanshen formula have more obvious roles which were mentioned before. It prove that The action of Shouwudanshen formula connect with the dose.
