A high performance liquid chromatography(HPLC) method was developed todetermine the liensinine.The conditions was as follows,Hypersil ODS2 C18column(4.6mm × 250mm,5um);UV detective wavelength 283nm;flow rate0.8ml/min;sensitivity 0.01AUFS;mobile phase is composed of methanol:pH7.2phosphoric acid aqueous solution (71:29) for in vitro samples and blood samples.Bywhich the interference of impurity may be avoided and there is a good linear relationshipbetween concentration and peak area. Three solvent methods were researched to extract total phenolic alkaloids,the bestis the methaol-acid solvent-ether method. Solubility,UV absorption,LogP,stability,pKa,plasma protein binding rate ofliensinine were determinted respectively,the results indicated liensinine solvent issensitive to light and temperature, plasma protein binding rate is low,determination ofLogP is 6.24. The permeation of total phenolic alkaloids of SEMEN NELUMBINIS throughhumen hypertrophic scars skin in the presence of several permeation enhancers wasperformanced with pH7.2PBS and modified Franz-diffusion cells for the first time.Theresults show that 3%Azone and 5%Propanediol together used may enhance thepenetration of total phenolic alkaloids to the highest degree.The research of deposit effectshow that 80% Liensinine was distributed in scars tissue and 20% Liensinine permeatescars tissue.
