Objective:To investigate the effect and mechanism of Compound Traditional Chinese Medicine Unguent on the hypertrophic scar(HS)in rabbit ears.Methods:The experiment was completed in the Laboratory of Department of Plastic Surgery,Xijing Hospital of the Fourth Military Medical University from July to December in 2006.â‘ 16 New Zealand rabbits(3~4 months year old)were chosen as experiment animals.â‘¡The models of HS in ears were reproduced.All rabbits were classified into 4 groups randomly:the experiment group(treated by the compoud traditional Chinese medicine ointment),the medicineâ… control group(treated by contractubex),the medicineâ…¡control group(treated by asiaticoside)and the blank control group.There were eight ears(forty-eight wounds)in each group.When epithelized,the wounds were treated respectively by the compoud traditional Chinese medicine ointment,contractubex,and asiaticoside three times per day, except that the blank group was treated by nothing.â‘¢On the 30thday and the 100thday four specimens were taken from each rabbit.And then the appearance and pathology were observed;the scar thickness and Hypertrophic Index(HI), blood flow of micro-circulation,collagen,a-smooth muscle actin and transforming growth factor-betal were tested;fibroblasts number was count.â‘£T test was used in the comparison of two groups;variance analysis was used in the comparison of multitude groups.Results:All the ears of 16 rabbits entered the result analysis.OThe quantity of HS in the experiment group is fewer than that in the medicineâ…¡group and the blank group,but more than the medicineâ… group.â‘¡Blood flow of micro-circulation was higher significantly in the experiment group than that in the control groups(P<0.05).â‘¢Long-term curative effect of the compoud traditional Chinese medicine ointment is conspicuous.â‘£Fibroblast counting on the 30thday was obviously lower in the experiment group than that in the blank group(P<0.05);on both of the 30thand 100thday,expression of a-smooth muscle actin was obviously lower in the experiment group than that in the blank group(P<0.05).⑤Quantitative determination and semi-determinalion of collagen on the 30thand 100thboth were lower in the experiment group than that in the blank group(P<0.05).6 Expression of transforming growth factor-betal on the 30th day was obviously lower in the experiment group than that in the blank group(P<0.05).Conclusions:The compoud traditional Chinese medicine ointment can inhibit the formation of the hypertrophic scars of rabbit's ears,lessen the degree of fibrosis of hypertrophic scar,reduce the fibroblasts and inhibit the hypertrophic scar contracture. |
