Rhodebryum roseum (Hedw) Limp has been used to treat the cardiac disease whose syndrome is mainly heartache, flustered, cardiopalmus and short breath for hundreds of years. In order to find activity compounds from Rhodebryum roseum (Hedw) Limp, The EtOAc extracts were systematically separated with various chromatographic techniques. 12 compounds were isolated and identified. Their structures and the stereochemistry of some compounds were determined by means of various modern spectroscopic techniques and chemical methods.These compounds were: uridine(l), ursolic(2), 3β-hydroxy- 5α,8α - epidi -oxygosta - 6,22- diene (3), 3β- hydroxy-5α,8α-epidioxygosta-6,9 (11) ,22-triene(4), caffeic acid methyl ester(5), methyl piperate(6), piperate(including 3 stereoisomeride, 7, 8, 9), β-sitosterol(10), daucosterol(11) and diphenoethene (12). Diphenoethene was a new compound with novel skeleton. Others excep t P-sitosterol were obtained from Rhodebryum roseum (Hedw) Limp fort he first time.The different anti-myocardial ischemia activities of water, 50% EtOH, 95%EtOH extractions of the Rhodebryum roseum were investigated by different models. The decline of S-T segment in ECG was lightened and the levels of serum GOT, LDH, CK were decreased in all groups administrated with Rhodebryum roseum. Compared with the controls, the 50% EtOH extract high dosage group was significantly different with the controls in S-T segment, LDH and CK(P<0.05). Myocardial MDA content was significantly depressed (p<0.05) and myocardial SOD was higher than those of the controls in the 50% EtOH extract higher dosage group.Low-dose of EtOAc and n-Butanol fractions obviously reduced idence rate of ventricular fibrillation, delayed the appearance of arrhythmia in mice and significantly prolonged the survival time of mice under the normal pressure and hypoxia. And they could also obviously prolong the living time of mice... |
