Tripterygium wilfordii Hook f. is a traditional Chinese medicine and is widely used in China. The extracts of Tripterygium wilfordii have various activities, including anti-inflammatory, anti-tumor, immunosuppressive and gonadal injury. Clinically, it was used to treat rheumatoid arthritis, systemic lupus erythematosus, chronic nephritis and other autoimmune diseases.In our previous studies, it has been reported several kinds of components such as sesquiterpene alkaloids, diterpenes, triterpences, lignins and other compounds. In this paper, one new sesquiterpene alkaloid (wilfornine H) and one new diterpene (triptobenzene Q), as well as 10 known compounds (ebenifoline E-II (3), 3-deoxy-maytol (4), sugiol (5),2-phenanthrenol (6),tripterifordin (7), triptophenolide (8), triptonoterpene (9), triptoquinone H(10),triptinin A(11),triptonide(12)) in Tripterygium wilfordii were elucidated by using spectroscopic methods('H-NMR, 13C-NMR,'H-'H COSY, HSQC, HMBC, NOESY) and high-resolution mass spectrometry.For finding immunosuppressive agents from the exact of T. wilfordii, we examined the inhibitory effects of the isolated compounds on lymphocyte transformation. Compounds 9-11 revealed a significant distinction compared to the concanavalin(ConA) control group (P<0.01,n=6), and showed inhibitory effect on lymphocyte transformation by comparing with a reference compound (Dexamethasone).Compound 12 revealed a strong inhibitory effect and its IC50 value is 44.72 ng/mL.Rhodobryum roseum Limpr. is a folk traditional Chinese medicine, which is widely used in Yunnan and other provinces. The extracts of Rhodobryum roseum Limpr. have various activities, including anti-myocardial ischemia, anti-arrhythmic, anti-atherosclerosis, anti hypoxia and anti-lipid peroxidation. Clinically,it was used to treat ischemia heart disease, nervous breakdown, diabetic retinopathy and other diseases.It has been reported terpenes, steroids, flavonoids, alkaloids, and other components isolated from this plant.In order to search the bioactive constituents from Rhodobryum roseum Limpr.,we try to isolated compounds from R. roseum Limpr. using kinds of column chromatographies, including normal phase chromatography, reverse phase chromatography, gel chromatography, and high speed counter current chromatography (HSCCC).Besides, we used GC-MS analysis to disclose components in this plant.7 known compounds (dehydrovomifoliol(1),mono-ethyl fumarate (2),4-hydroxybenzaldehyde (3),4-hydroxybenzoic acid (4), p-coumaric acid (5), vanillic acid (6), ursolic acid (7)) were elucidated by using spectroscopic methods (1H-NMR,13C-NMR,1H-1H COSY, HSQC, HMBC, NOESY) and high-resolution mass spectrometry.To investigate the cardioprotective effects and relative mechanism of R. roseum Limpr.,we cultured myocardial cells of neonatal rats, which were exposed to hypoxia-reoxygenation (H/R) injury, and detected the activity of lactate dehydrogenase (LDH), malondialdehyde (MDA), and superoxide dismutase (SOD). The results revealed a significant distinction compared to the (H/R) injury group and control group (P<0.01,n=6),which demonstrated cultured myocardial cells of neonatal rats were protected by R. roseum Limpr. from injury of hypoxia-reoxygenation (H/R).
