[objective] To investigate the protective effect of Astragalus Membranaceus onischemia-reperfusion injury in rat kidneys and its influence on renal cell apoptosis andits possible mechanism in protecting kidneys with ischemia-reperfusion injury.[methods] Thirty Wistar rats were randomly divided into 3 groups: control group,ischemia-reperfusion group and Astragalus Membranaceus group. Ischemic renal injuryin rats was made by placing the atraumatic vascular clamp in renal arteries for45minutes. Group A (ischemia-reperfusion group),in which ischemia was made by tyingboth renal arteries for 45 min,followed by 24-hour reperfusion;group B (AstragalusMembranaceus group),in which Astragalus Membranaceus was applied intravenously 10min before blocking of bilateral renal arteries for 45 min, followed by 24-hourreperfusion .The equal volume of normal saline was given to ischemia-reperfusiongroup ;Group C ( control group), in which sham operation was performed;The levels ofserum creatinine, blood urea nitrogen, plasma superoxide dismutase (SOD),malondialdehyde (MDA)and the influence of Astragalus Membranaceus on cell apoptosiswere observed when 45 minutes ischemia and 24 hours reperfusion were completed.[results] The levels of plasma BUN.Cr.SOD and MDA in the AstragalusMembranaceus group were (9.40 ±2.60) mmol/L , (100.42 ±4.02) μmol/L,(241.69±19.00 )U/ml and (6.206 ± 0.599)μmol/ml ,respectively and those in theischemia-reperfusion group were (14.25 ±2.13) mmol/L, (276.67 ±3.18) nmol/L,( 170.45± 14.27)U/ml and (8.998 ± 0.345)umol/ml .respectively. The levels of SOD in theAstragalus Membranaceus group were significantly higher than those in theischemia-reperfusion group (P<0.05), but the levels of MDA ,BUN,Cr in the AstragalusMembranaceus group were significantly lower than those in the ischemia-reperfusiongroup (P<0.05) .The indexes of renal cell apoptosis in the Astragalus Membranaceusgroup , the ischemia-reperfusion group and sham operation group were (15.6 ± 3.5) ,(28.5±4.4), (3.10 ± 2.2),respectively. Astragalus Membranaceus group were significantlylower than those in the ischemia-reperfusion group (P<0.05).
