Objective To investigate the inhibitory effects of glycosaminoglycan from scallop skirt (SS-GAG) on the foam cell induced by Ox-LDL and to discuss its mechanisms of anti-atherosclerosis.Methods 1.The porcine artery smooth muscle cells (SMCs) were incubated with 15mg/L ox-LDL for 72h to establish a smooth muscle cell-derived foam cell model. Intracellular total cholesterol (TC), free cholesterol (FC), cholesteryl ester (CE) content and CE/TC ratio) were determined through enzyme way to study the effects on foam cell formation.2. In vitro it took a model of SMC foam cells induced by ox-LDL, some chemical methods including xanthine oxidase method and TBARs were used to test the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX).3. In vitro it took a model of SMC foam cells induced by ox-LDL, some chemical methods including nitrate reductase method were used to observe the secretion of nitric oxide (NO).4. U937 cells were incubated with 80mg/L oxidized low-density lipoprotein (ox-LDL) for 48h :o establish a macrophage-derived foam cell model in vitro.
