Objective To investigate antivirual effects and its mechanism of Glycosaminoglycan from scallop shirt(SS-GAG) in vitro and in vivo.Methods 1.In vitro experiment:①With Vero cell as host cell,the influences of SS-GAG on reproduction and adsorption and synthesis of herpes simplex virus typeⅠ(HSV-Ⅰ)were investigated in vitro by MTT colorimetry and observing cytopathic effect. The cytoxicity of SS-GAG and the time-effect relationship of its anti-viraleffect were measured by MTT colorimetry.②With Hela cell as host cell,the influences of SS-GAG on reproduction and adsorption and synthesis of herpes simplex virus typeⅠ(HSV-Ⅰ) were stutied in vitro by MTT colorimetry and observing cytopathic effect.And the cytoxicity of SS-GAG were measured by MTT colorimetry.2.In vivo experiments:The mice models infected with HSV-Ⅰwere established by full-inoculation through abdominal cavity of mice to observe the influences of SS-GAG on the survival time,life prolongation rate and mortality rate.And the influences of SS-GAG on their pathological changes were observed by histological methord in mice infected with HSV-Ⅰ.3.The influences of SS-GAG on immune function:The effects of SS-GAG on the phagocytic activity of peritoneal macrophage and the proliferation of splenic lymphocyte were stutied by MTT colorimetry in mice infected with HSV-Ⅰ.The index of spleen and thymus were determined to study the influences of SS-GAG on function of the immune organs.Results 1.In vitro experiment:①The TCID50 of HSV-Ⅰwas 10-4.7.And the SS-GAG did not shown toxicity on vero cell within the concentrations of 25-1600μg/ml. Compared with the virus control group,SS-GAG(100μg/ml,50μg/ml,25μg/ml)could protect the Vero cells infected by HSV-Ⅰ,increased proliferated activity,and inhibited the cytopathic effect and the reproduction and adsorption of virus(P<0.01).These effects became stronger with the increase of concentration of SS-GAG..But the direct inhibiting effect of SS-GAG on the virus was not observed.After the 24~72 hours-treatment, SS-GAG could protect the Vero cells and heighten the cells proliferated activity(P<0.01 ). ②The SS-GAG did not shown toxicity on Hela cell within the concentrations of 50-800μg/ml.Compared with the virus control group,SS-GAG(200μg/ml,100μg/ml,50μg/ml) could protect the Hela cells effectively and heighten the cells proliferated activity.And SS-GAG could lessen the cytopathic effect induced by HSV-Ⅰand inhibit virus replication and intrusion(P<0.05,P<0.01 ).But SS-GAG was not observed to have a direct inhibiting effect to the virus.2.In vivo experiments:Compared with the virus control group,the SS-GAG(10mg/kg,20mg/kg,40mg/kg ) significantly lengthened the survival time and decreased mortality rate and lighten the pathological damage of some important organs in the mice infected with HSV-Ⅰ(P<0.01).3.The influences of SS-GAG on immune function:SS-GAG could significantly enhance phagocytic activity of peritoneal macrophage of mice infected with HSV-Ⅰand promote proliferation of splenic lymphocyte induced by concanavalin A and lipopolysaccharide (P<0.01).Meanwhile,the index of thymus of mice treated by SS-GAG improved significantly(P<0.01)Conclusion The glycosaminoglycan from scallop shirt(SS-GAG) has depressant effects on HSV-Ⅰin vitro and in vivo,and the mechanism of anti-virus effects attribute to improve function of the immune organs and the inhibit virus replication and intrusion.
