Part 1 THE EFFECT OF ESTROGEN ON THE HYPOXICPULMONARY VASCULAR REMODELINGOBJECTIVE: To observe the effect of estrogen on the hypoxic pulmonary vascular remodeling. METHODS:The animal model of hypoxic pulmonary hypertension was established by regular exposing the rats to normabaric hypoxic conditions for 3 weeks. Thirty male SD rats were randomly divided into five groups: control group, E2 preventive group, E2 preventive control group, E2 treatment group, E2 treatment control group. The mean pulmonary artery press (mPAP) was examined by right cardiac catheterization, and the right ventricle (RV) and left ventricle plus septum (LV + S) of rat were weighed respectively, and the RV/(LV + S) was calculated. Lung speciments were examined by light microscopes and morphological analysis semiquantitatively. The WT% ( ratio of vascular wall thickness/vascular external diameter) and WA% ( ratio of vascular wall area/total vascular area) was measured by a computerized image analyzer. E2 in serum was measured by means of the radio-immunoassay. RESULTS:1. In the E2 preventive control group, E2 treatment control group, mPAP, the RV/(LV + S), WT% and WA% increased significantly compared with those in the control group respectively(P < 0.01). Meanwhile, the serum level of E2 in hypoxic rats was not significantly different from that in control rats. 2. In the preventive and treatment group, the mPAP ,the RV/(LV + S), WT% and WA% decreased remarkably as compared with those in their control group respectively(P < 0.01). The serum level of E2 in the preventive and treatment group was higher than that in the control group respectively(P < 0.01). CONCLUSION:17-β estradiol may have the preventive and therapeutic effects on the hypoxic pulmonary vascular remodeling.
