The therapeutic effects of acupuncture on the reproductive endocrine disturbances of women and female animals have been reported. Acupuncture has been effectively used to cure some clinical gynecological sicknesses, such as polycystic ovary syndrome(PCOS), climacteric syndrome and other disorders of ovulatory dysfunction. However, the biochemical mechanism of EA normalization is unknown yet. And further probing the biochemical and molecular mechanism of EA normalizing the dysfunction of HPOA is not studied quite clearly yet. Our study tries to further probing the biochemical and molecular mechanism of EA normalizing the dysfunction of HPOA through the experiments by means of RIA, enzyme-radioassay, Immunocytochemistry and In Situ Hybridization.The experiment with ovarietomized rat as the animal model of menopause:1.SD rats were randomly divided into four groups: intact group (INT), intact group with electroacupuncture treatment (INT+EA); ovariectomized group (OVX) and ovariectomized group with electroacupuncture treatment (OVX+EA). Investigate the effects of electroacupuncture of "Shenshu" points, "Pishu" points on the blood levels of testosterone(T), estradiol(E2) and Progesterne(P) in female mouse by the means of RIA, and the effects on the ER by Immunocytochemistry.The result of the experiment prove that electroacupuncture "Shenshu", "Pishu" points of ovariectomized female rats can raise the blood levels of E2 and reduce the blood levels of T and ER; it has no evident affection of the levels of P, E2, ER and T of intact rats.2.The same method but with the "Shenmen" points, "Sanyinjiao" points and "Guanyuan" points. And the second experiment result quite met the first one. Result shows that electroacupuncture "Shenmen" points, "Sanyinjiao" points and "Guanyuan" points raised the blood levels of E2 and reduce the blood levels of T and ER; it has no evident affection of the levels of P, E2, ER and T of Intact rats.The experiment with 18-month-old female rat as the Animal model of climacteric syndrome:SD rats were randomly divided into four groups : intact group (INT), intact group with electroacupuncture treatment (INT+EA); 18-month-old aged female rat(AGE), 18-month-old aged female rat with electroacupuncture treatment(AGE+EA). Investigate the effects of electroacupuncture at "Shenmen", "Sanyinjiao" and "Guanyuan" on the blood levels of testosterone(T), estradiol(E2) and Progesterne(P) in female mouse by the means of RIA, the effects on the ER of hypothaiamus by Immunocytochemistry means, the effects on ERmRNA of hypothaiamus by In Situ Hybridization, and the effect on the aromatase enzyme activity of fat tissue by RIA. The result of the experiment prove thatelectroacupuncture "Shenmen" points, "sanyinjiao" points and "Guanyuan" points of old female rats can raise the blood levels of E2, P and reduce the blood levels of T, and it reduce the ER, ERmRNA and the aromatase activity of fat tissue; it has no evident affection of the blood levels of P, E2 and T of young rats.Conclusion: The experiments shows that the electroacupunture on the selected points actually compensate the reduced blood levels of. We presume from results that its the machnism lies on the raised aromatase activity. And the result of P, T blood leves and ER, ERmRNA of hypothaiamus could support the idea from some point and will provide experimental matierals for the further study on the machnism. |
