Objective 1. To observe the expression of HSPs in epidermal keratinocytes of psoriatic lesions and chronic eczama lesions .2. to investigate the role of HSPs in the pathogenesis of psoriasis and chronic eczama.3. to investigate the role of HSPs in identifying psoriasis and chronic eczama.Methods Expr-ession of HSP27 , HSP60and HSP70 protein and mRNA was detected by immunohistoch-emical SABC method and in situ hybridization in epidermal keratinocytes of psoriatic lesional skin in 33 cases psoriatic patients, chronic eczama lesional skin in 28 cases chronic eczama patients and 16 cases healthy controls.Results 1 .the quatity of HSP27 protein expressed in normal skin ,chronic eczema lesional skin and psoriatic lesional skin are 0.8919+0.9035, 1.0166+0.4869, 0.4852+0.5013 .In epidermis of psortic lesions,compared with the normal skin and chronic eczema lesions,the contents of HSP27 protein was obviously ruduced,the difference was highly significant(p<0.05,p<0.01),but in epidermis of chronic eczema lesional skin ,compared with the normal skin,the contents of HSP27 protein was not significantly different (p>0.05);The expressing tendence and the difference of HSP27mRNA was consistent with its protein.2.There was high expression of HSP60 in keratinocytes of psoriatic skin,but little and no expression of HSP60 in normal skin and chronic eczema lesional skin.The expression of HSP60mRNA was consistent with its protein.3. the quatity of HSP70 protein expressed in normal skin ,chronic eczema lesional skin and psoriatic lesional skin are 0.9251 +0.6721, 1.0313+0.5288, 0.8577+0.6266. But the difference of HSP70 protein between psoriatic lesional skin, chronic eczema lesional skin and the normal skin was not significantly different (p>0.05). HSP70mRNA was consistent with its protein. There has another result was that the expression of HSP27,HSP60,HSP70 and its mRNA has no significance in the progressive stage and resting stage of psorasis.Coclusion 1.The abnormal expression of HSP27and HSP60 significiently correlated with the pathogenesis of psorasis. HSP70 may play a certain role in psoriatic cecular protective mechanism.2.HSP27 and HSP70 may play a certain role in chronic eczama stress protective mechanism.3. HSP27 and HSP60 may be help to identify psorasis and chronic eczama in clinic and in pathology.
