Effects Of Chronic Stress On Serum Heat Shock Protein 70 And Corticosterone Contents Of Rats, Intervention Of Shen Qi Wan

Research purposeThe research of whole adaptability of Chronic stress is one part of research method on whole in physiology.When body is stimulated,the mean changes in nerves and endocrine system are the stimulation in LC-sympathetico-adrenomedullary system and HPA as well as the changes of many other endocrine hormones.HSPs are stress proteins which are very stenoplastic on evolution,they reside in every living thing from bacterium to human.HSP70 is the richest group in HSPs.HSP70 participates the process of folding,assembling,dispelling and reparation of proteins,regulates the activity of proteins,mediates the translation of protein,protects the self-steady system of proteins.In common-it is very little in most of histiocyte,but when body incurs different physiological,pathological or psychological stresses,HSPs will express more.In the 1^st experiment,we choose healthy rats as research object,and do not destroy their organism integrality.We give 3 types of chronic stresses:they are fitting swimming which represents suitable physiological stress,restriction which has the typical character of physically and mentally stress and the combination of such both.We determinc the HSP70 and corticosterone in rats serum when they have been stressed,observe and analyze the different of HSP70 which is entirety proteinum marker in order to study how fitting swimming,chronic restrict stress and the combination of such both to affect on body on the level of molecule-protein.In the 2^nd experiment,we put healthy rats under the stress of chronic restriction,and observe the intervention of Chinese medicinal materials-shenqiwan to chronic stress.Research methodThe 1^st experiment:1.Experiment animal:48 mature male Wister rats,weight(270±10)g.2.Experiment groups:All rats are divided into 6 groups which are control group, swimming group,restriction group,and combination group.3.Preparation of animal model:Swimming group:Practice fitting swimming without any weight loading every day for 4 wecks continually.Restriction group:Give one restriction stress every day for 4 weeks continually.4.Detection of HSP70 and Corticosterone of rats' serum:Feed rats of control group 4 weeks,stressed rats of each experiment groups 4 weeks,decapitate,recipe, centrifugalizate,abruption serum after stress in 24 hours.Use ELISA kit to detect.5.Statistics method:Experimental data is demonstrated by((?)±s).Analyze analysis of variance with statistics soft spss16.0.The 2^nd experiment:1.Experiment animal:48 mature male Wister rats,weight(270±10)g.2.Experiment groups:All rats are divided into 6 groups which are control group, restriction group,Chinese crude drug and Chinese crude drug control group.3.Experiment medicine:Chinese crude drug prescription is Shenqiwan from Jinguiyaolve,Chinese crude drug control is Gancao.4.Intragastric administration:The rats of Chinese crude drug and Chinese crude drug control group were administrated intragastricly with Shenqiwan and Gancao differently 5ml 1hr before restriction stress every day,continue 2 weeks.5.Detection of HSPT0 and Corticosterone of rats' serum:The same as the 1^st experiment.6.Statistics method:The same as the 1^st experiment.Finding1.The 1^st experiment:The HSP70 and corticosteronc in experiment groups rats' serum are higher than control group.It has statistical significance(P＜0.05).There is interaction between swimming and restriction.The HSP70 and corticosterone in rat serum are different and have statistical significance between each two experiment groups(P＜0.05).2.The 2^nd experiment:The HSP70 and corticosterone in rat serum of restriction group and Chinese crude drug control group heighten,which has statistical significance(P＜0.05),The HSP70 and corticosterone in rat serum is different and has statistical significance(P＜0.05) between Chinese crude drug group and Chinese crude drug control group or Chinese crude drug group and restriction group.But between Chinese crude drug control group and restriction group the difference has no statistical significance.(P＞0.05).Conclusion1.The 1^st experiment:Swimming can neutralizate the effect of restriction and lessen the harmful effects of restriction stress to body.2.The 2^nd experiment:Chinese crude drug Shenqiwan can lessen the harmful effects of restriction stress to body.
