Ets proteion constitute a family and function as transcription factors. As a member of Ets, ETS1 plays important roles in differentiation, lymphoid cell development, invasiveness and angiogenesis. Such diverse roles of ETS1 are likely to be dependent on its associated proteins. Here we describe a novel ETS1 interacting protein designated as ETS1-associated protein II (EAPII). Objective: On the levels of gene and protein, we studied the expression of ETS1 and EAPII in various cervical lesions. Especially,the function mechanism of ETS1 and EAPII during the devepments of cervical cancer. In the same time, we preliminarily studied that EAPII modulates ETS1 transcriational activity. In order to provide the theories basis for clinical diagnosis of precancerous and earlier carcinomas. Method: In this study, the technology and method are based on molecular biology. First, ETS1 and EAPII genes were amplified by RT-PCR After bait being generated. the result showed that EAPII and ETS1 was inbihited each other on mRNA levels. Following nuclear extracts were prepared from cervix and western blotting analysis indicated the expression of ETS1 in various cervical lesions on protein levels. At last, labeled with 32p-ATP, Electrophoretic mobility shift assay(EMSA) were utilized to detect the binding of protein and DNA. To determine whether EAPII regulates the DNA-binding abilty of ETS1. Result: In this paper, we significantly identified the relationship between ETS1 and invasiveness of cancer through investigating the expression of ETS1 and EAPII.The result of RT-PCR indated that the expression of ETS1 increased following the development of cervical lesions. p<0.05. Also, the expression was higher in cervical cancer with lymphectasia. p<0.01. but the expression of EAPII decressed following the development of cervical lesions. p<0.05. It showed that EAPII negatively modulates ETS1 transcriptional activety. The result of Western-blotting was the same as it of RT-PCR. And EMSA showed the DNA-binding abilty of ETS1 increased following the development of cervical lesions.Also EAPII inhibited it. Therefore, EAPII is a novel ETS1 modulator that regulates specific aspects of the ETS1 functions.
