Objectives (1)To observe the histomorphologic development of human fetal hippocampus. (2)To observe the differentiated, growing and developed changes of the neurons in hippocampus. (3)To study the expression of autocrine motility factor (AMF) and autocrine motility factor receptor(AMFR) during the development of human fetal hippocampus. (4)To investigate the meaning and relationship of AMF and AMFR in fatal hippocampus and the development of hippocampus. Methods Twenty-nine human fetal hippocampus(11 week-full term newborn infant )were collected. According to characteristic of the hippocampus structure changes, group one include seven cases from 11th~16th week, group two include seventeen cases from 17th~28th week, group three include five cases from 29th~newborn infant. The intermediate piece of hippocampus were fixed, embedded and cut.(1)Hematoxylin-eosin(HE) staining observed the histomorphologic development of human fetus hippocampus.(2)Nissl staining observed the development changes of the neuronal tigroid body in hippocampus.(3)The immunohistochemical SABC staining detected the expression of neuron specific enolase(NSE) , AMF and AMFR. Results (1)In tissue section with HE staining ,from 11th to 16th week, the hippocampus exterior shape of fetus came into being. The distribution cell was to concentrated in hippocampus, the most of which are small and round. The CA1, CA2, CA3 and CA4 of hippocampus and dentate gyrus were differentiated after the 17 weeks . Hippocampus included polymorphic layer, pyramidal layer and molecular layer. When the size of cells increased, pyramidal layer could be seen pyramidal cells near to pyramidal layer. Dentate gyrus included molecular layer, granular layer and polymorphic layer. There are round and elliptical cells in granular layer. As the hippocampus progressed, the density of the cell of the gradually decreased, round cells became pyramidal, most of cells were medium size in pyramidal layer. After 29 week, neurons gradually mature, the size of cell increasing and the density of cell decreased in hippocampus.(2)Thionine staining, before 16th week, there isn't nissl body in fetal hippocampus.
