Objective To study the effect of cor pulmonale patient's serum on synthesis and damage of DNA in cultured rabbit pulmonary artery endothelial cells (RPAECs) and production of LDH, NO, ET secreted by RPAECs. In order to help the correlative research on cor pulmonale.Methods To evaluate the effect of cor pulmonale patient's serum on RPAECs in different doses and possible ways by which it exerts harm mechanism. To refer to Jaffe's methods and improve it, we estabilished a model of cultured rabbit pulmonary artery endothelial cells and obtain RPAECs, then it was digested by jiaoyuanmeil. Observe the form the endothelial cells. Immunofluorescence technique identified the EC. the fourth to sixth generation endothelial cell was selected to experiment. RPAECs incubated with cor pulmonale patient's serum in different concentration about 12h, MTT bise and Comet assay technique were used to check and measure the effect of RPAECs on synthesis and damage of DNA in RPAECs. Though ultraiolet continuous detection method, radioimmunoassay method, nitrate reductase method, LDH, NO, ET were used to evaluate the changes of the function of RPAECs's damage and secretion.Results 1. MTT metabolic rate: MTT metabolic rate of model was much lower than normal. Some endothelia cells even diedo The more higher rate of cor pulmonale patient's serum , the less influence of RPAECs metabolism, the lower speed and less quantity of DNA synthesizing. 2. One-celled gel electrophoresis: round phenomenon reveal from the RPAEsDNA electrophoresis delineation which incubated by 10% nomal serum.
