Danggui blood-enriching liquid was created by Li Gao , which is composed by Danggui and Huangqi equaling 5:1. Its main ruction is supplying a deficiency and enriching blood. It is one of the famous blood-enriching compounds and is used publicly in clinic. Now, there are many researches on its use for pharmacology, but there is not report on its pharmacokinetics, which limits the use in clinic a certain extent. This dissertation systemically studied the pharmacokinetics of Danggui blood-enriching liquid for the first time and did a shallow study on the distribution of Ferulic Acid ( FA ) ,which is an important active component in Danggui blood-enriching liquid in tissues of rats and the rule of compound.Firstly, a study on the extraction process of Danggui blood-enriching liquid was carried out by a L9 (34) orthogonal experiment. The concentration of Ferulic Acid was as the target of the experiment. The best extraction process was as following: reflux with water for 3 time, and 12 times water was used for 0.5 hours in each time.The system of acetonitrile-triethy lamine was used to detect the concentration of Ferulic Acid for analysis inside for the first time. We developed an reversed -phase high performance liquid chromatographic(RP-HPLC) method for determination of FA in rat serum and tissues. The calibration curves of FA in serum and different tissues were created and got the linear range of every curve. The recovery coefficient and the coefficient of variation in serum and tissues were examined. The RP-HPLC method was simple, sensitive and accurate that was developed in this work and used to determined serum drug concertration by external standard method. It would provide a scientific basis for the pharmacokinetic study of Danggui blood-enriching liquid.The pharmacokinetic study was that blood samples and tissue samples were collected before and at various time after ig a single dose of Danggui blood-enriching liquid in rats. Then, prepared the sample of demand and determined them by RP-HPLC. The concerntration-time curve was analyzed with 3P87 program todetermine the compartment model and pharmacokinetic parameters. The test proved that concentration-time curve of FA in serum was fitted to two-compartment model. The main pharmacokinetic parameters were showed as following: ka = 0.2791 ±0.0321 min"1, ti/2ka=2.4838± 0.0633 min, ti/2a=3.4041±0.4325min, ti/2p =225.2843+25.3642 min, AUC=9368.7441 ±2145.7824 ng.ml/'.min"1. The result proved that Danggui blood-enriching liquid was fitted to dose as liquid.Ferulic Acid could be determined in heart, liver, lung, kidney. The concentration of Ferulic Acid was different in tissues. There was more drug in liver and kidney. Liver was the highest concentration in three time. The result showed that Danggui blood-enriching liquid was accumulated in the liver and offered some evidence for the traditional theory of science of Chinese meteria medica.We changed Danggui and Huangqi equaling 1:1, and got the pharmacokinetic parameters and the situation of distribution in tissues. Comparing with 1:1, drug could keep in the serum for long time and higher concentration, and had a better absorbtion when Danggui and Huangqi equaling 1:5.Thinking the distribution in tissues, at 1:1, drug was accumulated in the kidney in a short time and maybe poisoned body a certain extent. For the above reason, we offered some evidences for the rule of compound.
