Objective To investigate a more effective way to enhance the effect of irradiation of hypoxic human cervical cancer line(HeLa cells) by hyperthemia and CMNa.METHODS 1.The experiment was performed with HeLa cells in vitro, set up HeLa Hypoxic Cell Model, took count of the colonies by the standard in vitro colon formation methods. Irradiation with oxygenated HeLa Cell was control group, Irradiation with Hypoxic HeLa Cell was experimental group, treated with various X-ray schedules, drew the cell-survival curves, and calculated each group'D0, OER. 2.To test the cytotoxicty of CMNa. Drew the cell-survival curves after the hypoxic cells exposed to CMNa at several different concentrations for two hours by the cell culture technique, got the formulation, calculated the LD50. 3. To test the radiosensitivity of HeLa cell dealt with 0. 24mmol/l, 0.48 mmol/1, 1.44 mmol/1 consistency CMNa . Irradiation alone was control group, treated with various X-ray schedules, drew the cell-survival curves by the cell culture technique, calculated each group'D0, SER. 4. To compare the sensitivity between hypoxic cells and aerobic cells to hyperthermia.Hypoxic cells were experiment groups, aerobic cells were control groups. Drew a family of survival curves exposed to water bath for various periods of time at several different temperatures, compared the sensitivity between hypoxic cells and aerobic cells by SAS8.0.5.Hyperthermia after or before irradiation were experimental groups, irradiation alone was control group, treated with various X-ray schedules, drew the cell-survival curves, and calculated each group's Do, TER. 6.Then choose the optimal group: irradiation with high consistency CMNa and hyperthemia (given after irradiation), compared with irradiation alone group, drew the cell-survival curves, and calculated its SER.Result l.The hypoxic cell model was set up successfully, OER was 2.76, 2.The formulation is lgY= -0.0952-0.0099* , LD50 is20.79mmol/L, under the concentration about 4. 8mmol/l, CMNa had not the cytotoxicty to hypoxic cells, F=0.89, P = 0.468> 0.05. 3. The SER which the CMNa consistency was 0.24mmol/l, 0.48mmol/l, 1.44mmol/l was 1.57, 1.67, 2.28 respectively. 4.Hypoxic cells and aerobic cells have the similar sensitivity to hyperthermia. Under 4TC, 42°C, 43°C, P is 0.07, 0.10, 0.08 respectively. 5.The group by 42°C water heated 1 hour before irradiation, TER was 1.80. The group by 42 °C water heated after irradiation, TER was 2.04. 6. 1.44mmol/l CNMA combined with 42°C water heated after irradiation group's SER was 2.61.Conclusion 1. CMNa can enhance the effect of irradiation to hypoxic cells remarkably. 2. Hypoxic cells and aerobic cells have the similar sensitivity to hyperthermia. 3.The effect of irradiation can be enhanced by hyperthemia, heat given after radiation has a better enhancement than heat given before radiation. 4.The enhancement will better when they were used together.
