Objective This study was to explore the potentialfunction of Fas,Bcl-2 in the apoptosis of nuclei pulposus.Methods (1)The fresh pulpy nucleus tissue was obtained surgically and werefixed in 10%formalin , imbedded in Paraffin and sliced by 4μm.(2)The degeneration ofpulpy nucleus tissue was observed with HE staining. (3)In 20 cases of degenerativelumbar intervertebal disc tissues and 6 cases of normal lumbar intervertebal disctissues,the apoptotic ataus and the expression of Fas and Bcl-2 were detected withTdT-mediated JUTP-biotin nick and labeling(TUNEL) and immunohistochemistrymethods. (4) The results were analysed with SPSS 12.0 software.Results We detected apoptotic chondrocyte in lumbar intervertebaldisc.The average apoptotic index(AI) in 20 cases of degenerative lumbarintervertebal disc was 50.230,which was significantly higher than that of 6normal cases(28.317) (P<0.05).The expressions of Fasand Bcl-2 protein were in thechondrocytes of lumbar intervertebal disc tissues.The expression rates of Fasand Bcl-2protein in 20 cases of degenerative lumbar intervertebal disc were 74.255% and55.370%,respectively,which were significantly higher than those of 6 normalcases(41.717% and 27.167%, P<0.05).Conclusion (1)The high expression of Fasmay promote the apoptosis ofnuclei pulposus and the degenaration of intervertebral discs (2)The high expression ofBcl-2 may inhibite the apoptosis of nuclei pulposus and the degenaration ofintervertebral discs ,which may result from the harmful stimulation in the exterior and interiorenvironment of the intervertebral discs. |
