[Objective] To establish the model of unexplained spontaneous abortion in vitro, and explore the immunological mechanism or pregnancy immune tolerance and unexplained spontaneous abortion(USA).To provide theoretical foundation for effective immunotherapy of USA.[Methods] ① Harvest the supernatant from cultured immunological cells of deciduas from normal early pregnancy women and patients with unexplained spontaneous abortion, which were both stimulated by two kinds of trophoblast antigens. ②Concentrations of Interleukine-2(IL-2), Interferon- γ (IFN- γ ),Interleukine—4(IL-4)> Interleukine—10(IL-10) were determined by enzyme linked immunoadsorbent assay (ELISA). [Result] (DA11 the supernatant of cultured immunological cells of deciduas from normal early pregnancy have high level of IL-4 -. IL-10 and low level of IL^ IFN- v than in supernatant of USA. ?The concentrations of IFN- y >. IL-2 in supernatant of patients with unexplained spontaneous abortion were significant high than in supernatant of normal early pregnancy. [Conclusion] The disorder of immune cells of deciduas immunosuppressive ' s function is the main reason of unexplained spontaneous abortion. The immunological microenvironment of deciduas from normal pregnancy is dominant with Th2 type response. However enhanced Thl type response can mediate abortion.
