Serological Screening Cervical Carcinoma Associatied Antigens From CDNA Expression Library

Objective: Cervical cancer is the second most common carcinoma of gynic malignant tumor . It is estimated that more than 500,000 new cases of cervical carcinoma be diagnosed , and More than 190,000 women die of it each year . total livability of cervical carcinoma was 66% in developed country , but less than 50% in developing country . To explore some new immunotherapeutic molecules from cervical cancinoma , in this trial we screening cDNA expression library of hela cell line ( cervical carcinoma ) to find new tumor antigens by using SEREX .Materials and Methods: cDAN expression library of hela cell line ( Cervical cancer ) were bought from Stratagene Company , Goat anti-human IgG AP were bought from Sigma Company . cervical Sera were obtained routine diagnostic and untreated cases during August 2002 ~ Octuber 2003 from the Second Huaxi hospital of Sichuan University, and were stored at -80℃ or in liquid nitrogen until used . serological screening cDNA expression library of Hela cell-line by using SEREX ,strong clone.we obtained were cutting , sequencing and analysing with DNASIS and BLAST softwares in EMBL and GenBank.Results: (1) By using SEREX technology, 10 positive clones were obtained after immunoscreening, and 10 of 15 the nucleotide sequences of cDNA inserts were determined and analysed with DNASIS and BLAST softwares in EMBL and GenBank.Conclusions: These genes can be classified to 4 categorise: 1) tumor antigene ,for MU-MB-20.201. 2) cell modulation factor , for ABCA7. 3) structure gene: formin, AP3M1, et al 4) there are 4 clone we fined their sequences in human's gene bank , but not fined same protein . we may be obtain novel genes.
