Objective To investigate whether any change of STR genotypes occurred in recipients' blood collected at the different intervals after blood transfusion with different quantity of donors' blood Methods Three patients were transfused with 400ml, 800ml and 1 200ml blood respectively. The blood samples were collected either from the recipients' pretransfusion or at 4h, 8h and 12h after blood transfusion as well as the donors' blood and were anticoagulated with EDTA.DNAs were extracted by Chelex-100 method and were were stored at 4℃. Six loci of D1S549, D18S865, D3S1754, D12S391, D12S375, D6S477 were selected.DNAs were amplified by the polymerase chain reaction (PCR). The PCR products were analyzed by polyacrylamide gel (PAG) vertical electrophoresis and silver staining. Results The outcome of our research shows that the patients' six STR genotypes completely were consistent both in pretransfusion blood and in posttransfusion blood collected within 12h posttransfusion. Conclusion The STR genotypes of the donors probably have no influence on the patients' genotypes within 12h posttransfusion.
