Objective: To investigate the effects of rhein on apoptosis induced byTNF-α and TG in hunman hepatocyte line L-02,and to elucidate its possiblemechanism. Methods: The hepatocyte was cultivated in vitro.The hepatocyteinjuryed model of hunman line L-02 was induced by TNF-α(500u/ml)andTG ( 20ug/ml ) .Cell viability and proliferation was assayed byMTT.Flowcytometer was used for the hepatocelluar apoptosis.Bcl-2 andBax expression of hunman hepatocyte line L-02 were detected byimmunohistochemistry. Results: Compared with the control group ,RH has no effect onhepatocyte proliferation below the concentration at 5ug/ml.Both theconcentration of RH at 1ug/ml and 5ug/ml can block up the inhibitioryeffect of TNF-α and TG on hepatocyte.The hepatocellular apoptosisinduced by TNF-α and TG was significantly blocked up by RH(1ug/ml).Inthe RH group ,levels of Bcl-2 was up-regulated and Bax expression wasdown-regulated,compared with the control group both had significantdifferent. Conclusion: RH may have inhibitory activity on apoptosis of L02 cellinduced by TNF-α and TG.The mechanism of the effect may due toup-regulate the levels of Bcl-2 expression and down-regulate the levels ofBax expression.
