ObjectiveTo explore the mechanism of Astragalus Polysaccharide (APS) combined with Rheinon chronic renal failure(CRF) by studying the two drugs’ effect on endoplasmicreticulum stress(ERS)-induced apoptosis.Methods37male Wistar rats were randomly divided into normal group(N group), modelgroup(M group), low-dose group of APS (200mg/kg﹒d) combined with Rhein(25mg/kg﹒d)(L group), high-dose group of APS (400mg/kg﹒d) combined withRhein (100mg/kg﹒d)(H group). Wstar rats were fed with adenine (200mg/kg﹒d)by gastric tube to establish CRF model. After7weeks of treatment, renal function,serum electrolyte concentrations and pathological changes of all groups wereexamined, and Western-blot method was used to detect the expression of C/EBPhomologous protein(CHOP), inositol-requiring protein-1(IRE1),c-Jun-NH2-terminal kinase(JNK)in renal tissues.Results1. Kidney weight and kidney index: compared with N group, Kidney weight andkidney index of M group increased (P<0.01); compared with M group, Kidney weightand kidney index of L and H group decreased significantly (P<0.01).2. Biochemical parameters: compared with N group, BUN and SCr value of M groupincreased (P<0.01); compared with M group, BUN and SCr values of L and H groupwere significantly reduced (P<0.01); compared with N group, concentration ofcalcium ion of M group decreased (P<0.05); compared with M group, concentrationof calcium ion of H group was higher (P<0.05); compared with N group, phosphorusconcentration in serum of M group increased (P<0.01); compared with M group,phosphorus concentration in serum of L and H group decreased significantly(P<0.01). 3. Effect on renal tubular apoptosis: compared with N group, amount of apoptosiscells in renal tubular of M group increased (P<0.01); compared with M group, amountof apoptosis cells in renal tubular of L and H group were reduce (P<0.01).4. Protein expression of CHOP,IRE1and JNK: compared with N group, proteinexpression of CHOP,IRE1and JNK of M group increased (P<0.05); compared withM group, protein expression of CHOP,IRE1and JNK of L and H group decreased(P<0.05).ConclusionAPS combined with Rhein may alleviate renal cell apoptosis induced by reaction ofERS through reducing CHOP, IRE1, JNK expression in the rats with CRF, and thusplay a protective role for kidney.
