| [Objective ] To study the effection and mechanism of mouse islet transplantation on I type diabetes .[Methods] Using STZ make Balb/c and C57bl/6 mice become diabete model. Collagenase digested pancreatic tissue, islet cells were transplanted under the kidney capsula. Blood glucose level and immune cell number changes with flow cytometry method and local pancreatic tissue damage and apoptotic genes change with immunohistochemistry methods were compared after and before transplantation.[Results] Firstly, diabet model mice general states were improved and its blood glucose went normal and diabete symptoms and objective signs were relieved after islet transplantation. Compared diabete mice with treatment groups, inflammation reaction were lightened in transplanted mice pancreas . Secondly, the number of CD4 positive and CD8 positive cell in centre and peripheral immune organ have recovered after islet transplantation in diabete mice, which suggest mouse immune function gradually recover normal level after transplantation. Thirdly, pancreatic tissue damage were lessened, and its apoptosis related genes P53, Bax and Fas-L etc. expression decreased after transplantation in diabete mice. Fourth, islet cell transplantation partly inflammatory reaction were light after transplantation. Which suggest diabete treatment effects of islet transplantation is caused by islet transplantation and diabete immune function recovery is not caused by graft rejection reaction.[Conclusion] Mouse islet transplantation have significant treatment effect on diabet, its mechanisms probably have some concerned with immune system function change and pancreas cell apoptosis and inflammation reaction were lightened. |
