Objective To explore genetic and environmental risk factors of systemic lupus erythematosus(SLE) and to analyze the effect of environmental factors and interaction between environmental factors and CXCR4 and CCR3 gene in SLE. To study the factors affecting the therapy outcome of SLE and to provide indexes which can predict curative effect and prognosis of SLE so as to raise survival rate of SLE patients.Methods Genetic variance of CXCR4 and CCR3 were determined by PCR-SSCP. The environmental risk factors of SLE were analyzed by multivariate unconditional logistic regression and interaction between environmental factors and CXCR4 and CCR3 gene in SLE was analyZed by generalized linear model.To analyze clinical character , treatment result and prognosis of SLE with univariate and multivariate logistic regression analysis.Results The variance rate of CXCR4 gene amplification fragment in patients with SLE was 8.09%, in control was 7.33%, which has no significant difference (P>0.05). The variance rate of CCR3 gene enlarge section in patients with SLE was 19.85%, in control was 24.00% , which has no significant difference (P>0.05). Environmental risk factors(damp, psychic stimulation , sunlight, taking irritants food , taking celery,, infection of tonsil, times of miscarriage, and taking birth control pill for a long time) were selected to enter the logistic model.Result of generalized linear model showed that interaction between CXCR4 and CCR3 gene and environmental was irrelevant to pathogenic of SLE.
