BackgroundAluminum (Al) is the third most abundant element in the Earth's crust, humans are exposed to aluminum, from various therapeutic treatments and environmental sources, such as cookware, cans, some foods and drinking water.Aluminum is neurotoxicin that include dialysis syndrome , amyotrophic lateral sclerosis , and anemia is considered a possible etiologic factor in Alzheimer's disease and other neurological disorders, the metabolism and toxicity of Al have been study intensively in animals and humans.But, up to now , The molecular mechanism of aluminum-induced impairment of neurological functions remains unclear, So, the studies of the neurotoxic effects of Al are an urgent need .ObjectiveTo explore the sensitive neurobehavior indexes of subchronic administration aluminum mice by a series methods of behavior,the results can be referenced in early toxicosis in human. To study aluminum concentration in the mice brain regions,and lipid peroxidation in the brain,meanwhile, observe the levels of intracellular calcium, DNA damage in brain cell was observed by comet test. To find toxicity mechanism of Al and provide scientific basis for revising intake limit criteria.MethodsAfter Subchronic exposured to Al,observe learning and memory ability in mice by Morris water maze and olfactory discrimination task, spontaneous motor activity by spontaneous motor apparatus , wire suspension and balance beam test detect balancecapability ,anxiety by Black-white alley and Plus maze, Species-typical behaviors by Hoarding and Nest construction .aluminum concentration determined with ICP, levels of SOD and MDA in brain were measured individually with test boxes ,free intracellular calcium was determined in synaptosomes with the calcium-sensitive indicator dye fura-2/AM,DNAdamage was measured by single-cell gel electrophoresis, the degree of DNA damage was indexed by the percentage of cell with comet-like tail. The data were analyzed with SPSS 10.0Results(1) After Subchronic exposured to Al in mice, we observe breath and behavior of mice, which were normal, body weight were no significant (^359=0.937, P >0.05) , anxiety behavior were no significant in four groups.(2) Compared with the control, The times of spontaneous activity of Low dosage group, medium dosage group and high dosage group were lower than those of control respectively (/><0.01), and correlated inversely with dose and the times of spontaneous activity(r=-0.421 .P<0.01).Grip strength scores of medium dosage group and high dosage group scores were lower than those in control scores, (P < 0.05).Transform scores of beam traversal and tight rope task were not significant in all groups.(3) Spatial learning and memory ability was showed no significant (F=0.202 P— 0.894) , non-spatial learning and memory ability in olfactory discrimination task, medium dosage group and high dosage group performed worse than control respectively (/=5.59 P=0.018) , (/=4.92 ?=0.0265)o(4) Aluminum increased food hoarding from a source outside the home base, food hoarding was weightier in high dosage group than that in the control,( ^=8.407 P= 0.038).
