Oxytetracycline(OTC) and metacycline(MC) are the kind of better antibiotic pharmaceuticals, their antibacterial spectrum are as same as that of the tetracycline but their curative effects are superior than that of the tetracycline, they can also be used to the therapy of urethra and respiratory tract infection. And because of their wide antibacterial spectrum and high potency, Oxytetracycline and metacycline are common antibiotics used against bacterial infections. Adenosine disodium triphosphate (ATP) is the mediator of energy exchanges that occur in all living cells, both catabolic, degradative processes and anabolic, biosynthesis processes. Because of its ubiquitous presence in living matter, ATP has been widely used as an index for biomass determinations in clinical microbiology, food quality control and environmental analyses. The nicotinamide adenine dinucleotide phosphate (NADP) is natural coenzymes present in all animal and plant organisms. They are essential to the function of many catabolic and biosynthetic pathways found within living organisms. Each of these forms has a particular role in enzyme regulation, during differentiation and similarity processes. Nicotinamide adenine dinucleotide phosphate has a pyridine circle of nicotinamide, and using it oxygenation and deoxidization can occur reversibly, it can be used for the transfer of hydrogen in the metabolizability. One of the appreciated roles of the nicotinamide adenine dinucleotide phosphate is in the prevention and/or reversal of oxidant damage within the cell. Therefore, it is significant to establish a precise, sensitive and simple method for the determination of NADP. In this paper, according to above three objects, using molecule spectroscopy (fluorescence and Uv-Vis spectroscopy), we study the interaction between drug-metal ion complex and biomolecule and its application. There are four chapters. First, I summarized my thesis and introduced the investigation actuality of fremdness and homeland. Second, we synthesized DE-β-CD with a non-polar cavity to provide a protective micro-environment for guest molecules (OTC/MC-Eu3+) to form the inclusion complex to gain the further enhancement of sensitized fluorescence of Eu3+ ion by preventing fluorescence from quenching. Experimental results indicate that the characteristic peak of Eu3+ ion at 612nm can be more greatly enhanced in OTC/MC-Eu3+ system by DE-β-CD. According to these, a new fluorescence method with high sensitivity and selectivity is established for the determination of OTC or MC. Third, using rare metal ion-drug complex as fluorecence probe, study the interaction between the complex and adenosine disodium triphosphate (ATP) and its application, this drug including doxycycline, doxycycline and ciprofloxacin. After addition ATP to Eu3+-drug system, the characteristic fluorescence intensity of Eu3+ or Tb3+ could be enhanced greatly, and the enhanced fluorescence intensity is in proportion to the concentration of ATP. According to this, we constructed a new method for the determination of ATP. The developed method was practical, simple, sensitive and relative free from interference coexisting substances and had been successfully applied to the determination of ATP in samples. Fourth, there is energy transfer between doxycycline and Eu3+ and the complex can emit the characteristic fluorescence of Eu3+, so the complex can be used to determine nicotinamide adenine dinucleotide phosphate (NADP) as a kind of fluorescent probe.
