In order to discover novel antibiotics dealing with antibiotics-resistant bacteria, we establish in vitro model of screening anti-MRSA and anti-VRSA. We screened 3000 strains in which 70 strains showed positive including strain SIPI-434. A compound named SIPI-JY-1 from the broth of strain SIPI-434 was isolated and by analyses of its physico-chemical qualities, UV, MS and NMR spectra etc., the structure of SIPI-JY-1 was elucidated and is identical with that of genistein.As reported before, strain SIPI-100 could produce antitumor compound SIPI-100-1 and other compounds so that we wish to find novel antitumor compounds. By the optimization of fermentation conditions, scale-up of fermentation, solvent extraction and silica gel column chromatography, another active compound named SIPI-100-2 was isolated;by its physico-chemical qualities, UV, IR, MS and NMR spectra data, its structure was elucidated and is identical with that of 17-O-demethylgeldanamycin. In the activity study, SIPI-100-2 showed strong antitumor activity.Both strains SIPI-434 and SIPI-100 were thought to belong to genus streptomyces through the study of their morphologies, culture properties and physiological-biochemical properties. |
