Objective: 1. To study the injury of hippocampal neurons after the kindled epilepsia models of rats induced by Coriaria Lactone (CL). 2. To study the different changes of Calcitonin gene-related peptide (CGRP) in hippocampus and dentate gyms of the kindled epilepsia models of rats induced by Coriaria Lactone (CL), and to explore the possible effect and action mechanism of CGRP in pathogenesis of epilepsy. Methods: 30 healthy male Sprague-Dawley (SD) rats, weighing between 80g-100g, were randomized into test group (n=24) and control group (n=6). The test group received repeated CL intramuscular injection at subthreshold dosage (0.2ml/kg) until the rats were kindled, and the control group received normal saline (NS). These injections repeated every 60hours. After average injection for 18 times, the rats whose seizures reach 3-5grade (Racine scale) within 24 hours after CL injection and have 10 or more seizures between 2 injections were thought as the kindled rats. The rats that do not reach the above conditions were thought as the non-kindled ones. The brain tissue ofall groups (kindled group, non-kindled group and control group) were dissected and collected in order to perform HE and Amino-Toluene-Blue stain. On the other hand, the cry-sections of brain tissue were immunohistochemically stained, then were examined and analyzed semi-quantitively for expression of CGRP by means of Image-pro plus4.5 digital micro-image analysis systems. Results: (1)11 rats were kindled. The kindled rats had all grade (Racine scale) seizures after CL injection. The pathology change of brain tissue was observed with HE stain, hippocampal neurons loss was observed with Amino-Toluene-Blue stain. (2 ) CGRP of the kindled rats in hippocampus and dentate gyms increased significantly as compared with that of the non-kindled rats and the control rats (p<0.05). Conclusion: 1.There was obviously brain neurons injury of the kindled rats induced by CL. 2. CGRP of the kindled rats in hippocampus and dentate gyms increased significantly as compared with that of the non-kindled rats and the control rats, and CGRP maybe play an important role in epilepsy.
