Objective The aim of study is to investigate the effects and mechanism ofbreviscapine and antisense sequences of thrombin receptor on vascular smooth muscle cells proliferation induced by thrombin.Methods The fourth-sixth generation Sprague-Dawley rat arteriole vascular smooth muscle cells which cultivated by adhesive cultivation was to be applyed, Cells was treated by antisense thrombin receptor oligo-deoxy-nucleotides and difference density of breviscapine solution (0.5ug/ml 5ug/ml 50ug/ml) , a negative control-group was established without any treated. The effects of breviscapine and antisense sequences of thrombin receptor on rat vascular smooth muscle cells proliferation under thrombin was observed by using 3H-thymidine amount which act as the index of cells proliferation degree. The effects of breviscapine and antisense sequences of thrombin receptor on thrombin receptor mRNA level was observed by reverse transcription polymerase chain reaction( RT-PCR) method.Results Compared with control-group, breviscapine and antisense thrombinreceptor oligo-deoxy-nucleotides inhibited significantly the proliferation of Sprague-Dawley rat arteriole vascular smooth muscle cells induced by thrombin, there were statistic difference among them (P<0.05),but breviscapine-group has almost thesame inhibitory degree on cells proliferation as well as antisense thrombin receptor oligo-deoxy-nucleotides group in the trial. RT-PCR assay showed that breviscapine and antisense thrombin receptor oligo-deoxy-nucleotides restrained markedly thrombin receptor mRNA expression much than control-group whose expression was 2-3 times as much as amount of the formers. There are no difference between breviscapine group and antisense thrombin receptor oligo-deoxy-nucleotides group.Conclusions Vascular smooth muscle cells proliferation induced by thrombin canbe inhibited by breviscapine and antisense sequences of thrombin receptor, breviscapine and antisense sequences of thrombin receptor arrest vascular smooth muscle cells thrombm receptor mRNA expression. The restrained thrombin receptor gene expression may contribute to the explanation of one of the mechanism of the inhibited vascular smooth muscle cells proliferation.
