Objective: To set up the methods for the quality control of Rubia yunnanensis (Franch.) Diels and Indigo Naturalis. Content: 1. To determinate To develope an analytical method for determination of rubiquinone in Rubia yunnanensis by RP-HPLC. 2. To establish a method for the determination of free anthraquinone in Rubia yunnanensis (Franch.) Diels. 3. To develope a simple high-performance liquid chromatogxaphy(HPLC) method for separation and quantitative determination of indigotin and indirubin from Indigo Naturalis. 4. To study the extracting technology condition of indirubin in Indigo Naturalis. Results and Conclusion: 1. A new RP-HPLC method has been established. It is accurate, steady, and with good reproducibility. 2. A new chromatoptometry has been established. It is quick, simple, accurate and with good reproducibility, and suitable for determination of free anthraquinone in Rubia yunnanensis (Franch.) Diels. 3. A selective HPLC method to simultaneously determinate isomeride (indigotin and indirubin) was developed and validated. It could be adapted for quality control of Indigo Naturalis. 4. Simple factor analysis experiments and orthogonal test method were used for studying the effect of different extracting factors (the differet solvent, the extracting time, the solvent volume, the temperature and so on) on extracting rate of indirubin. The process is reasonable and favorable for industrial production.
