Objective: Our study was to investigate the relationship between hMSH2 and p16~INK4a protein expression and explore the correlation of hMSH2 and p16~INK4a protein expression with lymph node metastasis and prognosis in gastric cancer by using tissue microarray(TMA) of gastric cancer and look for the molecule marker to predict prognosis in gastric cancer.Methods: TMA of gastric cancer was formed to explore expression of hMSH2 and 16~INK4a in 120 gastric cancer by immunohistochemical methods(IHC). All data was processed by using SPSS 12.0 software, x~2 test and Fisher exact test were used to investigate the relationship between hMSH2 and p16~INK4a protein expression. Kaplan-Meyer Log-rank method and Cox multivariate analysis were used to explore the correlation of hMSH2 and p16~INK4a protein expression with survival. For all statistical test, a P values less than 0.05 was defined as statistically significant. Results:1, In TMAs of gastric cancer(120 cases), 114 hMSH2 and 116 p16~INK4a were valuable. The usable rate of TMA(hMSH2) was 95.0% and the usable rate of TMA (p16~INK4a) was 96.7%. The usable rate of TMAs was over 95.0%.2, In 120 gastric cancer cases, the overexpression rates of hMSH2 and p16~INK4a proteins were 56.1 % (64/114) and 71.6% (83/116) . Apositive correlation was found between the overexpression of hMSH2 and p16~INK4a (r=0.23507, P=0.0134).3, In 120 gastric cancer cases, the overexpression of hMSH2 and p16~INK4a proteins were not related to sex, age, size, histology type respectively. The overexpression rates of p16~INK4a was related to infiltrative dept ( P<0.01 ) .4, In 114 hMSH2 and 116 p16~INK4a, the overexpression of hMSH2 and p16~INK4a proteins showed no significant association with lymph node metastasis, UICC N stage.5, In 120 gastric cancer cases, the patient with gastric cancer live 27 monthaveragely. The overexpression rates of hMSH2 and 16INK4a proteins were not related to prognosis (P=0.0617, P=0.7349 ) .Conclusions:1 ^ The development of TMA of gastric cancer was used to investigate hMSH2 and pjgiNK4a protejn expression for high-throughput analysis and make the result high standard and believable.2, The overexpression of hMSH2 and p^1**48 was closely involved in the carcinogenesis of gastric cancer.3 ^ A positive correlation was found between the overexpression of hMSH2 and ^s was SUpp0Se(i mat me overexpression of hMSH2 promote the expression4> In gastric cancer , hMSH2 could not predict lymph node metastasis ^ UICC N stage as a molecule marker. If hMSH2 was an independent prognostic factor or not need to be studied.5 > In gastric cancer , the overexpression rates of i6INK4a show significant association with infiltrative depth. There were not relationship between pl6INK4ao-verexpression and lymph node metastasisN UICC N stage> prognosis. The result did not support that pl61tiK4a overexpression is an independent prognostic factor.
