To study the effects of nimesulide ,a selective COX-2 inhibitor, on proliferation and apoptosis of human gastric carcinoma cells SGC-7901 in vitro, and provide an experimental basis for clinical application.Proliferation of human gastric carcinoma cells SGC-7901 was assessed by MTT ,and apoptosis was detected by flow cytometry.The experimental result showed that, Different dosage (0, 20, 50, 100, 200, 400μmol/L) Nimesulide has the inhibitory action to the human gastric cancer cell SGC-7901. 20μmol/L Nimesulide inhibitory rate is lowest, after different time (24h,48h, 72h, 96h) ,the inhibitory rate respectively is: 14.1 %, 20.3%, 21.8%, 29.3%;400μmol/L Nimesulide inhibitory rate is highest, after (24h, 48h, 72h, 96h) , the inhibitory rate respectively is: 29.3%th, 31.1%, 42.6%, 51.7%, 82.3%. At the same time Nimesulide changed the gastric cancer cell SGC-7901 cell cycle distribution, different density Nimesulide (0, 20, 50, 100, 200, 400μmol/L) affects for 72 hours, the G2/M cell accounts for the proportion respectively is: 10.4%th, 12.2%, 14.9%, 16.5%, 20.0%, 26.7%. After 100, 200, 400μmol/L Nimesulide affects, Flow cytometry demonstrates gastric cancer cell SGC-7901 appears obvious apoptosis rate respectively is: 17.7%th, 22.6%, 75. 2%.Nimesulide may inhibit the proliferation of gastric adenocarcinoma cells SGC-7901 remarkably due to the affecting the distribution of cell cycle and inducing apoptosis.
