ObjectiveSSRIs antidepressant drugs are frequently used drugs to treat depressive disorders. This study is to investigate the relationship between genetic polymorphism of 5 - hydroxytryptamine traneporter ( 5 - HTT) intron 2 variable number tandem repeat ( VNTR) and clinical response adverse reaction of selective serotonin reuptake inhibitors ( SSRIs ) .MethodsThe study consisted of 82 Chinese Han inpatients diagnosed as major depressive disorder. The patients are divided into two groups according to the therapeutic effect or adverse effect and to analyze the difference of the genetic polymorphism of 5 — hydroxytryptamine traneporter(5 - HTT) intron 2 variable number tandem repeat (VNTR). PCR Cycling Profile:94℃ , 4min;[94℃: , 1min - 60℃ , 45" - 72℃ , lmin] x 35cycles;12℃ ,10min. Genetype is divided by polyacrylamide gel electrophoresis.ResultsIn total, there were no significant differences of genotypic and allelic distribution of 5 - HTTVNTR between different effect of drug treatment (fluoxetine) , the frequency of genotype and allelic distribution in patients with adverse reaction of nausea is significant different with the frequency of genotypic and allelicdistribution of 5 — HTTVNTR in patients without this adverse reaction.ConclusionThe findings suggest that the 5 - HTTVNTR gene polymorphism may be associated with the adverse reaction (nausea) of fluoxetine. Future studies with larger sample sizes and other drugs of SSRIsare needed to confirm this finding.
