Midazolam is a new-type of Benzodiazepines medicine, it has been widely adopted in clinical anesthesia because of its advantages, such as short preclud half-life, anterogrucle amnesia and high reliability. Meanwhile it possess other virtues: strong sedation, anticonvulsion and antianxious.The study was designed to use amygdala kindling model in rats,to investigate the antiepileptic role of midazolam,and supply experiment fundation for reasonable medication in clinical.Aim To investigate the inhibition and antiepileptic effect of midazolam on amygdala kindling models in rats.Methods To establish amygdala kindling models in rats and determine the effect of different dose of midazolam on kindling models and its antiepileptic effect;to observe the effect of midazolam on independence activity of mice;convulsions were determined by maximalel electroshock seizure( MES )model in mice.Results Midazolam(0.25-1.0mg·kg~-1, ip) were shown to dose-dependently inhibit amygdala kindled seizure in rats (P<0.01), significantlyshorten the after discharg duration(ADD) and reduce the Racine's stage(P<0.01 );Midazolam(0.5-1.5mgkg~-1 ip) can significantly decrease theindependence activity number in mice ( P<0.01). Meanwhile, midazolam candose-dependently inhibit the maximalel electroshock seizure in mice( P<0.01), decrease convulsion number and convulsion ratio.Conclution Midazolam inhibits the seizure of the amygdala kindling , having significant antiepileptic effect and obvious anticonvulsion and sedetion effect , suppling experimental fundation for reasonable medication and developing new pharmacological value.
