| Objective To evaluate the anti-inflammatory potency of mizolastine objectively and exactly at the dose of anti-histamine efficacy and explore its possible mechanisms.Methods Thirty-six rats were divided into three groups randomly and twelve per group,the reaction of inflammatory edema in rat paw was induced by a single subcutataneous of arachidonic acid (1.0mg/ml, 0.1ml). Mizolastine (0.3mg/kg) ,cetirizine (0.3mg/kg) and isasteric normal sodium was given orally 2 hours before the injection.Results It is observed that mizolastine (0.3mg/kg) significantly inhibited arachidonic acid induced inflammatory cell infiltrate and mast cell degranulation on light microscope (P<0.05);Cetirizine (0.3mg/kg) has no significant inhibitory effects on arachidonic acid induced inflammatory cell infiltrate and mast cell degranulation (P>0.05) .Collecting foot pat of rat paws at 2h,4h,6h after the administration,and carring out histopathologic observation,we observe inflammatory cell infiltrate after HE staining and shape of mast cell after TB dyeing.The results were analyzed by statistical package for the social science 11.0(SPSS 11.0 FOR WINDOWS).Conclusions (1)The results suggest that mizolastine of antihistaminic dose has higher anti-inflammatory activity in vivo. (2) Histological observation find that mizolastine exert its anti-inflammatory effects by inhibiting infiltration of neutrophilic leukocyte and mast cell degranulation. |
