Eight new and nine known compounds were isolated from the xylem ofTripterygium wilfordii by using column chromatography, including MPLC. Theirstructures were elucidated on the basis of spectroscopic studies, including 1H-NMR,13C-NMR, 1H-1H COSY, HSQC, HMBC, NOESY, MS spectra, etc. Thesemisystematic name of new compounds were 1β-hydroxy-2β,5α,11-triacetoxy-7β-nicotinoyl-8β-benzoyl-dihydroagarofuran(1), and 1β,5α,11-triacetoxy-7β-nicotinoyl-8β-benzoyl-dihydroagarofuran (2), 1β-furanoyl-2β,3α,7α,8β,11-pentacetoxy-4α,5α-dihydroxy-dihydroagarofuran (3), 1β,2β,3α,5α,7β,8β,11-heptacetoxy-dihydroagarofuran (4), 1β-furanoyl-2β,3α,7α,8β,11-pentacetoxy-5α-hydroxy-dihydroagarofuran (5), 1β,7β,8α-triacetoxy-2β-furanoyl-4α-hydroxy-11-isobutyryloxy-dihydroagarofuran (6), 1β-nicotinoyl-2β,5α,7β-triacetoxy-4α-hydroxy-11-isobutyryloxy-8α-furanoyl-dihydroagarofuran (7), 2-ketone-1(5),6,9-guaiene (8).And nine known compounds, 1β,2β,5α-triacetoxy-4α-hydroxy-7β,11-diisobutyryloxy-8α-furanoyl-dihydroagarofuran (9), triptoquinone A (10), wilforic acid A (11),triptoquinone H (12), triptonoterpene (13), triptonodiol (14), triptoquinone B (15),triptobenzene H (16), hinokione (17), were isolated from the xylem of T. wilfordii forthe first time.In a screen for immunosuppressive agents from Tripterygium wilfordii, weexamined the inhibitory effect of isolated compounds on lymphocyte transformation.Through the experiment of lymphocyte transformation test, most constituents showeda significant distinction to the concanavalin (con A) control group (p<0.01, n = 6), andshowed inhibitory effect on lymphocyte transformation by comparing with a referencecompound (dexamethasone). Compounds 10-17 revealed strongerimmunosuppressive activities.
