[Objective] To explore the effect of the hepatic lipase gene promoters -250G/A and -514C/T polymorphism on plasma lipoproteins metabolism and coronary heart disease.[Methods] We studied 101 patients with CHD and 74 controls. Using the method of polymerase chain reaction - restricted fragments length polymorphism to determine the genetype of the hepatic lipase gene promoter -250G/A and -514C/T .Automated Biochemical Analyzer was used to measured the concentration of the plasma lipids.[Results] (l)The levels of plasma HDL was significant lower while TG, LDL and apoB were significant higher in CHD group than controls . (2)The C→514T substitution in the promoter region of the HL gene is associated with higher HDL , LDL, apoAI and apoB levels. G→250A substitution is associated with higher HDL and apoAI levels. When we analysis the subsititutions C→514T and G→250A together, our study has shown that 93 cases with two subistitutions, 72 cases without the two substitutions, 10 cases only with one substitution( -250G/A). HL -514T together with -250A alleles are associated with higher plasma HDL, TC and apoAI levels. (3) The frequencies of the -514T allele is 0. 371 in the control group and 0.257 in patients with CHD. The results are not all the same with foreign researches. The frequencies of the -250A allele is 0.385 in the control group and 0.307 in CHD group. No demestic research has been reported. (4)In multivariate logistic regression analysis, the -514T allele is a protect factor for CHD.[Conclusion] (l)The frequencies of -514T allele is siginificant lower in patients with CHD than that in the control group. The frequencies of -250A allele has no differences between two groups. (2)The -514T allele (CT and TT genotypes) is associated with higher plasma TC% HDLn LDL> apoAI and apoB levels. (3)The -250A allele (GA and AA genotypes) is associated with higher plasma HDL and apoAI levels. (4) The frequencies of the -514T allele is 0. 371 in the control group and 0.257 in patients with CHD. The frequencies of the -250A allele is 0. 385 in the controls and 0.307 in CHD group. (5) In multivariate logistic regression analysis, the -514T allele is a protect factor for CHD.
