Objective: To determine whether the expression of mRNA of bcl-2 and bax and the apoptotic lymphocytes in human SLE patients are abnormal by analyzing mRNA of bcl-2 and bax and inquire into the roles of which in the pathogenesis of SLE. Methods: The levels of the expression of mRNA of bcl-2 and bax of 41 SLE patients and control group of 32 age and sex-matched healthy donors were detected by RT-PCR;While anti-ENA autoantibodies of SLE patients were detected with Western-blot;and complement (C3) of the patients and the control group was tested quantitatively by turbidimetry. Results : 1 the levels of expression of mRNA of bcl-2 and bax in every group: â‘ The levels of expression of mRNA of bcl-2 of active SLE patients(n=27) was significantly higher than that of the quiescent SLE patients(n=14, P<0.01), and of the normal healthy donors(n=32, P<0.01),and that of quiescent SLE patients(n=14) was significantly higher than that of control groups(n=32, P<0.01);Of the active SLE patients, no difference was found between the incipient and the treated with corticosteroid or immunosuppressive therapy (P>0.05). â‘¡The levels of expression of mRNA in SLE-nephritis patients was significantly higher than that in non-SLE nephritis patients(P<0.01). 2 Statistically significant differences were shown between , one group with two and more kinds of autoantibodies, and another group which had at most one kind of autoantibody ( P<0.01) in SLE patitents;3 The levels of expression of mRNA of bax were no difference in every group. Conclusion: 1 The levels of mRNA of bcl-2 of SLE patients increase significantly,especially in active SLE patients and patients with more autoantibodies,so it should be confered that high level mRNA of bcl-2 perhaps palys an important role of occurance and development of human SLE. 2 The ratio of bcl-2/bax unbalances,so many lymphocytes can not apoptotic normal.which make their lives longer and result in some T lymphocytes and polyclonal B lymphocytes activated and proliferating overage in SLE patients, which perhaps is one of reasons of producing much autoantibodies. 3 High level PBMC bcl-2 mRNA in SLE patients hold back apoptosis of many lymphocytes ,assemble them on kidney tissue,which perhaps contribute to pathogenesis of lupus .
