ObjectiveTo evaluate the effect of Valsartan on endothelial function in patients with essential hypertension by measure the naitric oxide, superoxide dismutase, malondialdehyde in serum and endothelium-dependent dilation function in brachial artery. MethodsThere were 36 patients of essential hypertension in the research, take 80mg Valsartan daily. Blood pressure and the side effects were observed weekly. After 8 weeks, NO in serum were measured by way of naitric oxide deoxidized, SOD and MDA (i.e. LPO) by way of chemically colorimetric method, assessed brachial artery endothelium-dependent dilation function by way of high resolution extrinsic blood vessel ultrasound. The control group was consisting of 26 health adults. Result1 .The effect of Valsartan on underpressure The total effective rate of Valsartan to hypertension patients is 75%. The SBP of the test group before and after the treatment were (153.6±10.6)mmHg, (136.5±10.3)mmHg respectively. The DBP of the test group before and after the treatment were (102.1 ±6.8) mmHg, (90.2±4.9) mmHg respectively.2.Changes in brachial artery internal diameter Changes in brachial artery caused by reactive hyperaemia after treatment was significantly weakened in the group of essential hypertension Compared with the normal control, significant difference exited (P<0.01) and improved obviously after the treatment.3.Before taking Valsartan, statistic difference of NO level was observed between the group of essential hypertension and the normal control ( P <0.01) .After taking Valsartan, significant improvement was verified. ( P <0.01 ). 4.Prior to treatment, SOD activity of the group of essential hypertension was significantly lower than that of normal control ( P<0.05), MDA activity was increased ( P<0.05) . After the treatment of Valsartan, SOD activity increased greatly, significant difference was tested( P <0.05), and MDA activity decreased evidently ( P <0.05)? 5.Correlation analysis Single regression analysis verified the existence of linearity positive correlation between NO level and the changes in brachial artery caused by reactive hyperaemia after the treatment of Valsartan ( P <0.01) . Conclusion1.Without treatment, blood vessel endothelium function in essential hypertension patients decreased dramatically, NO ^ NO and SOD level were significantly lower than those of normal control while MDA level increased with statistical meaning. 2.After 8 week of the treatment by Valsartan, along with the obvious effect of underpressure, the improvement in the level of NO, SOD, as well as the decrease in MDA level, statistical significance was validated.3.Before treatment, changes in brachial artery internal diameter derived from reactive hyperaemia decreased greatly, significant difference exit with normal control (P <0.01), indicating endothelium-dependent dilation function. Mediated by blood flow in the group of essential hypertension was weakened and improved significantly after the treatment ( P <0.01), proving Valsartan can improve endothelium-dependent dilation function.
