Recent years, the most progress in cardiovascular field is certification that vascular endothelium plays an important roles in regulating heart circulation function and stabilizing internal environment . It's interpretation that vascular endothelium dysfunction is relating to occurrence and development of some cardiovascular disease. Therefore estimating the vascular endothelium function posses important clinical significance to cardiovascular disease 's prevention and cure. It is generally accepted that the hypercholesterolemia is considered independent risk factor to coronary heart disease. Endothelium dysfunction has an high incidence in hypercholesterolemia. Endothelium dysfunction is earlier period pathologic alteration of atherosclerosis and takes part in the whole progress of coronary heart disease occurrence and development. Through the drug ameliorated vascular endothelium function in hypercholesterolemia prevent atherosclerosis and coronary heart disease . Nitric Oxide ( NO) is the most important relaxation factor that regulating the vascular tension. Nitric Oxide Synthase ( NOS) is the only catalysis enzyme in vivo. To explore the influence of lowering lipid to the hypercholesterolemia, this study observe the change of the EDD, NO and NOS in hypercholesterolemia which have been therapied by fluvastatin. Its clear the relation that the lowering lipid effect of statin and amelioration of vascular endothelium.MaterialsPatients: A group of 35 patients with hypercholesterolemia in the outpatients from the first affiliated hospital of the China medical university in 2003 fit-ting the following criteria was selected for this study: total cholesterol(TC)>6 5mmol/L and/or low density lipoprotein cholesterol( LDL - C) >3. 4mmol/L, meanwhile , and/or triglycerides(TG) >2. 3mmol/L. 25 healthy adults who fitting the following criteria were as control group: TC < 5. 7mmol/L,TG < 2. 2mmol/L,LDL - C <3. 3 mmol/L,high density lipoprotein cholesterol( HDL -C) > 1. 1 mmol/L. Both the patients and the healthy adults who had others diseases such as CHD, severity heart failure, arrhythmia, diabetes mellitus, disease of thyroid gland, and had taken lowing lipid medicine within 3 weeks were not included in this study.Methods1. General observation marker(1) Asking the history such as smoking, hypertension, DM, CHD family history and physical inspection.(2) Chemical examination detected blood lipid, blood sugar, ECG,UCG and so on.2. Medicine in patients with hypercholesterolemia during the observation Patients with hypercholesterolemia were treated with fluvastatin (40 mg/d,po) for 8 weeks.3. Detecting blood lipidPatients with hypercholesterolemia detected blood lipid at 8-.00 -9:00 before treatment and after treatment 8 weeks. Enzyme technique was used to detect TG. Oxidase technique was used to detect TC, LDL - C, HDL - C. Blood lipids group was detected only once in the control group.4. Detected vascular endothelium function by ultrasound doppler Patients with hypercholesterolemia detected vascular endothelium functionbefore treatment and after treatment 8 weeks. Vascular endothelium function was detected only once in the control group. We use ultrasound doppler to detect vascular endothelium function. We detected the brachial artery bore's bottom value, reactive hyperemia, response to nitroglycerin.5. NO,NOS measurements2ml median cubital venous blood were collected in tubes right and were kept 30 min at the room temperature . It was centrifugated 10 min at 2000rpm/ min and the serum were collected. Then it was been kept at - 20C. Nitrate re-ductase technique was used to measure NO. According to the test kit instruction: blood serum normal reference value is 65. 7 + 15. 8 mol/L. Chemistry chromatometry was used to measure NOS. According to the test kit instruction: blood serum normal reference value is 37.84 +3.63U/ml.6. Statistical analysisAll statistics were conducted using SPSS11.0 statistical package.
